New, couple questions
    #8020187 - 02/13/08 10:42 PM (15 years, 11 months ago)

Hey everyone just saying hi, I'm new to this but everyone starts somewhere.

I've just inoculated 4 full pint jars using vermiculite and BRF. Now, having done my reading and being generally good in the lab, I am a strong believer in sterile technique. Since contamination is the primary cause of failed jars, here's my question: when you pressure cook the jars you leave Aluminum foil on loosely to prevent water dripping but to allow steam to flow somewhat (if I recall). After you inoculate the cake, though, the foil isn't sealed airtight (or I've somehow missed noticing the step in growing guides). Wouldn't this leave a path for airborne contamination through the holes in the lid? I suppose a better question to ask: Is there a reason to avoid sealing the foil airtight (after pressure cooking) to avoid contamination, or is it a good idea?

I'm using B+ spores from a friend's print, he is only a few months more experienced than I am. My room temperature tends to fluctuate between 74-78F so I just put the jars in a box somewhere. Hoping for success, thanks for any pointers.

I'm pretty new myself, but I can say that it seems best to give the cakes breathing room, meaning holes for air exchange. The myc is a living organism and will give off gases, and use gases, so it can't be totally air tight. It does sound like the air flow "could" introduce contams, but the concern is minimalized by leaving the fewest and smallest possible entry points.

Dont worry about how tight your foil is on during sterilization.

You have a dry verm layer... right?
If so, theres no need for foil after sterilization. your layer of vermiculite will be the barrier for contaminates entering your jar after inoculation.

I used to use tyvek on my pf jars to be sure.

tight on the aluminum foil. you dont want water getting in..

at least that's what i do..

and leave lids loose for gas exchanges yes.. and it does allow a path for airborne contams, but that's why you have a DRY verm layer atop your cake..

if your worried put some mircropore tape, i used it on my wbs jars and 19/20 came out fully colonized, 1 may have been bruised from shaking but i threw it out anyways. im pretty new too tho.

This was covered in an interesting thread. Louis Pasteur proved well over a hundred years ago that unless contaminated air is fanned upwards into a container, contaminants being heavier than air cannot travel upwards into a container. He proved this by having sterile, nutrient rich liquids open in swan necked flasks. Some of his original flasks are still around, uncontaminated to this day.

ok, it's been a week since inoculation and I just took a couple pics of the jars. first visible growth was seen about 3 days after inoculation.

If it's hard to tell in the pics, looking closely you can see that any black spots are just bits of verm. The foil lids are off in these pics to give a clear view, but I've normally had them resting lightly on top. The jars on the left were inoculated in 4 spots and the jars on the right in 3. Does this look about normal for pint jars after a week? Would removing the foil lids full time help the jars colonize faster?

I also realized that I may be out of town for a while when the cakes are ready to fruit. If a jar is fully colonized, does it need to be fruited immediately or is there a way to preserve it for a week or so?

Actually believe it or not (I am sure people will agree) you "should" let the jars go about 1 week after full colonization.

I meant after that week, which I had already factored into my timeframe for going out of town.

ya you'll be ok, just keep them in a dark place...

also, you should give them as much gas exchange as possible without risking contams.. so i say take the foil lids off.. you've got a good layer of dry verm, right? that should protect them.
