Thanks! that kinda makes sense.
In a nut shell you want to isolate before it has a chance to cross breed with every spore in the pile so you can breed it with itself and eliminate the cross breeding you'd have to eliminate an agar again anyway!
if you don't that isolate will never be as good as it could have been because it already was crossed with a couch potato.
5 shrooms for you sir for finally making that clear to me!
I see oysters the size of car tires on the horizon!!

Yep, sounds like you got it. No matter what though it seems that no matter how early you catch the myc to transfer to a new dish you are still going to pull a few sectors(spores that have started to grow out). From reading it seems like it takes a few transfers to get your isolate. I mean after all spores are microscopic so im sure you can imagine how many spores there are in even a pin head sized surface area. Knowing that the easiest way to reduce those problems is to go light on the amount of spore solution used or scraped from a print so you get as few spores as possible which will cut down on the amount of sectoring needed to be done.

RR has a few good posts Ive read through about strain isolation and if you have a few dollars to spend buy "the mushroom cultivator" by Paul Stamets. I got it for 12$ off amazon and the book has a good deal of info about agar and strain isolation. I'm still working through the book but so far its a good read.

Good Luck with those tire sized oysters
Keith

Read it(most of it anyway).
What a painfully slow read for me.
I guess I fell asleep for that part.
I sold it before I really got deep into this hobby, guess I should give it another try.
I remember something about dilution of spore solution so you get 1 or 2 spores per drop...must be in there somewhere.
I got diverted by Liquid cultures at about that point.
