Some problems with WBS or spores?
    #8004435 - 02/10/08 12:20 PM (15 years, 11 months ago)

So I've knocked up 20 quart jars of WBS, and so far I have only had one jar germinate and finish. I started from multispore, so this could have been a problem for me. Each jar recieved 1cc. It took probably a week and a half for little white fluffies to show, and then another week and a half for it to finish out.

I did several small variation in procedure to see if there would be a difference in colonization time. I tried picking out the "floaters" from the rinse, and left them in for another batch. I tried soaking with coffee and gypsum, and soaking with just plain water. I also tried simmering and steeping with seperate batches. I even added a litte bit of fine vermiculite to some of the jars to see how it would affect the colonization.

Now only one jar even showed signs of mycelium. In case you were wondering it was one where I picked out the "floaters", soaked with just plain water, added a little verm, and steeped. I've already done a G2G with it and they're coming along quite nicely.

The jars that had no signs of germination were opened approximatly 30 days later, and 3 of 19 smelled just awful.(I'm assuming sour rot, but its not too important) The rest smelled like cooked corn...almost good enough to eat.(lol, almost)

So my question is...is there any way of knowing why I'm having a germination problem? I've had the exact same problem with LC's...I'm almost afraid to try agar...but I've got some agar and plates coming tomorrow. Any suggestions?

Sounds like bacteria in too-wet jars. I think the verm you added to the successful jar soaked up the extra moisture and helped with the problem. Grain should be on the dry side when you prep it.

Were they too hot when you shot them? Did you pressure cook them properly?

If you can't handle jars, you shouldn't mess with agar. I can tell you right now from experience that you will just waste money.

Hmmm....the reason that your jars are colonizing slow may be due to the fact that you used only 1 cc of spore solution, but that is probably not the best explanation. Just a thought here, have you made absolutely sure that your thermometers are in good working condition? That may be why they take along time, perhaps temp is lower then you think. I had one fuck up on me the other day and didn't realize that my incubator was at 105 until I opened the lid and felt how hot it was.

Did you make the syringes yourself?
