fractional sterilization w/ a PC and FAE questions
    #7986602 - 02/06/08 01:00 PM (15 years, 11 months ago)

Sorry about combining two separate topics into one post, but these are both important for my upcoming grow.

During my last attempt, I had some wet spot contamination in every jar I prepared. You can see my thread here. In the thread, RR's advice was "After boiling the grains, you must put them in a filtered jar and pressure cook right away. Bacterial endospores, once germinated, form new endospores within hours. By putting them in the refrigerator for a day or two, you got a whole new bacterial bloom, some of which appear to have survived the pressure cooker.".

Here's the thing: I've heard that repeated sterilization(fractional sterilization) can work with less than ideal sterilizing equipment. I have a pressure cooker, but it's old, and I'm not too sure about it. It worked fine for my PF jars, but it never rocks and doesn't have a gage: when it heats up, steam starts pouring out of the center jiggler or whatever the hell it is. I don't know if it works at 10psi or whatever. I pressure cooked every jar for 2.5-3 hours. I was wondering what anyone thought about this idea for preparing rye or WBS:
rinse then soak grain for roughly 24 hours. Drain, rinse, then simmer for 20 minutes. Drain. Put in jars. Pressure cook for 90 minutes. Let sit for a day to allow endospores to germinate, then pressure cook for 3 hours. Would this increase my chances against those fucking bacillus endospores? Any personal experience?

For my fruiting set up, I'm planning on using a closet with some transparent tarp and setting up an enclosed space. I've read that coolmists hooked up outside the greenhouse wear out quickly, so I was wondering how feasible it would be to have the coolmist or ultrasonic humidifier sitting on the ground, underneath either some perlite or paper towl for the drip, with either slits in the tarp or a corner of the enclosure being open(ie, lifting up a corner of the tarp so there's a hole through which air can pass)?

There is no need for fractional if you have a PC, then run it for 60-120 minutes at 15 psi, then let cool to room temp before you open (24 hours).

2 hours tops with quarts IME.

Soak 12-24 max, IME.

The reason I'm asking though is because I'm not so sure about my PC's abilities and a 3 hour PCing session apparently wasn't enough to kill of the endospores in my last batch of rye. I suppose I could get a new PC, but I'm trying really hard to conserve cash at the moment.

Sounds like you're just missing the rocker valve, so all you really need is a new one of those...

Bear in mind, my diagnosis in the other thread was little more than shooting from the hip. The bacteria could just have easily been introduced at inoculation, or via poor filtering in your lids. Bacteria is in the air everywhere, so if your filters are not up to par, or if they get wet, bacteria can colonize right through them.

I'd start looking for a pressure cooker that you can trust though. . .
