hello.
on Jan 14 I scratched spores from a print I got from FSRE onto 3 petri dishes. 1 contaminated and the other didn't show any growth at all.
the third one has been doing fine. mycelium started growing after a couple of days. first in the spots where the needle touched the agar, then 3 or 4 more spots came several days later.

yesterday two tiny "eyes" formed in the middle of the oldest mycelium "field". they look like tiny tiny little droplets of fluid (not from condensation). they're a little greenish and it looks like you can see through them a little. is this a contamination or are they starting to pin?

also, none of the mycelium "fields" that started growing looked rhizomorphic to me (it is my first grow though..) and just were almost perfect circles.

the spots I'm talking about can be seen at about 10 o'clock.

please upload future pics to the shroomery.
most people wont even look at pics hosted on image shack because of spyware.

that aside, it looks to me like you should isolate away from the black dot thats forming.
dark spots in your agar dish are most likely contamination.
mushroom mycelium is nice and white.

sorry, and I will. (never heard anything about spyware on image shack)

it's not even close to as dark as it looks on the picture.
more of a seethroughy tiny emerald or something.

are any of the growth zones rhizomorphic? to me it doesn't seem so.

First, don't post image shack pictures because we won't open them. Second, you should transfer each and every growth that begins on agar as soon as it's seen. Never allow dishes to grow out like that from spores, because any contaminants that are on the print can grow, contaminating your culture. If each growth gets immediately transferred, some dishes will be pure mushroom mycelium, while others will be all contaminants and can be discarded.

Post the picture here and I'll take a look.

I didn't have any petri dishes ready to transfer from.
this was also the last "surviving" dish. I had 10 made of which 7 contaminated before even being inoculated (I was using a glovebox, but only had an oven to heat them). of the remaining 3, 1 contaminated and one didn't show any growth at all.

I have h2o2 now and will prepare more dishes with it as soon as I have the time to, which won't be in the next few days though. should I put the one in the fridge for now?


am I right about there not being any rhizomorphic growth recognizable?

if it is contaminated, how can this have happened, long after that specific spot was overgrown with healthy looking mycelium?

Rhizomorphic strands appear to be trying to form from the larger section at the 7 to 8 o'clock position. I see three or four growths that should go rhizo with a transfer or two. Get plastic pre-sterilized petri dishes. Glass is not suitable in my experience for mycology, since you can easily go through twenty or thirty dishes doing a single series of strain isolations. It costs more to clean and re-sterilize glass dishes than what the plastic ones cost.

first thing I really need to get is a pressure cooker.
before getting one all I can do anyways is staying on agar with the mycelium I get from that current dish and use peroxided agar in future dishes. Bought malt extract earlier on since I am never ever going to use dog food agar again. ever. the smell is just intolerable.

my plan is then, once I have a strain isolated, to create a LC from that. had the idea to add h2o2 to the LC and just throw in a piece of mycelium. would that work, or do I really need to get a PC first?
