Air in syringe--could it be bad?

If one were to accidentally pull back the plunger on a spore-filled syringe, filling it with air while in a non-sterile environment, and then proceed to shake the syringe to break up the spore clumps, would it be a true statement to say that that syringe is now totally fucked? Or could one, after realizing the stupidity of his/her mistake, push the plunger and expel the air bubble, thereby saving one's syringe and sanity?

It probably won't matter that much because it's a pretty small amount, but it's definately a vector of contamination. It's a lot better to pull sterile air out of a sterile jar or bag that's been autoclaved if you have to.

That wouldn't make sense that it would save the sterility, if a contam is in that air, it's likely to make contact with the spore solution and probably contaminate it. If it's fucked, it's fucked, but don't throw it away by any means. Make a test jar and try it after getting rid of the air I suggest. Some grain or brf/verm is a lot cheaper then a new syringe or making another syringe, so try a test jar.

There isn't likely to be enough contaminants in 1-2 cc of air to cause any problems.

In the future though, you can put the tip of the needle in the flame, and suck in hot air from the flame to get sterile air into the syringe.

The bad thing is you may or may not be fucked. Next time put the tip of the needle into a flame then draw the air in.

My friend worried about the same thing - my friend didn't have any problems, but that could have been due to chance, of course.

It'll only kill the ones on the very top, near the syringe -- doesn't really matter.  Remeber -- the air in a sterilized jar or bag is sterile -- no flame is needed.  It's up to you :wink:
