IMHO, There's no need to soak the needle with alcohol after it's already bean burnt. You are giving it time to access contaminants.

I work with bacteria on a pretty daily basis and a hot flame WILL provide some cubic ft of air space. Learned that in like freshman biology lab.

A lit candle? It will provide some, but probably not that much. I'm thinking more along the lines of a bunsen burner.

I thought the purpose of alcohol was to you know.. kill contams? Plus, wouldn't the needle be too hot that it'd burn the spores coming through it?

If you're using a spray bottle, like you should be, then what are you worried about evaporation for? In fact, I'd probably want it to evaporate faster just to hurry up. It's my belief that 70% is the norm since anything above 70% is really just a waste of ethanol (since 70% will have the same toxic effects as 100%).

Alcohol is not an air sanitizer, it's a surface disinfectant. Big Difference! Using alcohol as an air sanitizer will just make your room a waiting fireball.

Sanitize means to reduce the number of contaminants to a safe or relatively safe level as may be judged by public health requirements.
Disinfect means elimination of all recognized pathogenic microorganism but not necessarily all microbial forms.
Sterilize means the destruction of all microbical life by use of chemical or physical procedures.

Was your post directed at me? Because I wasn't trying to say alcohol is an air sanitizer. I just meant when I use alcohol I like to use a spray bottle to apply it to surfaces.

sometimes I just hold my breath.
If I just wanna grab a quick clone and spike it into a dish or jar I seriously just crank on the oven element, stick the blade on it and when it's red I just do it as smooth and fast as I can.
Split the shroom, cut the sample drop it in, shut it in....Breathe...
Right in the middle of my filthy kitchen.

you'd be suprised how often I've gotten away with it.

If I'm doing a lot of stuff I shut off the furnace around midnight and do it REAL early before anyone has had a chance to kick up any dust.
One time my wife turned on the vacuum while I was working....totally fucked everything I had open and every dish I tried to do after that.
I am working on another glove box even though I haven't had many problems (except the vacuum thing)

Even when I'm in the most sterile room which lab's don't even have but I don't even breathe...If I do..I'll either breathe out the window or into the fan of the laminar flow hood.

Glovebox with a window open so I can blow out. I sterilize it from cm to cm then I hold my breathe even after I brush/Mouthwash, Shave Head, Face, Shower, all the above in my sterile technique just 'better safe than sorry.

Know what I do? I pressure cook jars and whatever i put in them. i flame sterilize my needle, then wipe it with alcohol. inoculate. done. no bleach, no oust, no bullshit. Contamination has never been a problem for me. I don't even have a glovebox. I could give a rat's ass if I lose 1 or 2 jars to contam. Or even a bag, since wbs is cheap and having lc means I can do lots of innoculations. I simply cannot believe the things some people do.

good luck doing tissue cultures in a sterile room without breathing :laugh: i practically lived in a sterile room + working in a positive pressure hood and would spend hours in there over the summer. youre definitely overkill dumbfounded, but hey. if it works and you dont mind the extra caution then more props for ya

I recommend Ozium ... they pick that as the best Air Sanitizer out there....they say Ousts kills 99.9% .. BULL SHIT...Ousts is for Soccor Moms who wanna come home to a non smelly home.

Ozium is recommended for Hospital Sanitiation. Lab Work, Surgeries and has a finer mist! You use about 4-5 sprays and be as effective as using a half to a whole can of OUST! A 14.5 oz can will go for months! Trust me...try it out...I've never had a complaint with it and they recommend it now way better then OUST.
