From what I've read, consensus from the experienced with strain isolation is; potency comes mostly in part from genetics.
With my current knowledge and experience the only way I could conceivably isolate for potency would be to clone a fair amount of fruitbodies, producing several flushes (one of each clone) then injest multiple test samples over several days to determine which clone was most potent. Problems with this method are numerous. Not to mention time consuming.
Now, on to the questions.
Is there a faster way to achieve this task? Such as a way to measure alkaloid levels of individual fruitbodies after clones have been taken? Is such a venture even worth the tribulation? I don't want to waste my time if the outcome isn't a significant difference from your average specimen. But, saying that: the reason I started this thread is because I've fruited several trays in a single grow from the same multispore syringe and noticed a significant potency difference between trays, raising my curiosity. I don't like having to eat double doses for single does effects.

Perhaps take an equal mass sample for all your prospective clones and perform an alcohol extraction of the psylocibin. Then you could weigh the resulting crystal from each sample and see which has more alkaloids per mass

That sounds valid. I can't think of any extraneous factors that could mess you up that way.

You could use a cheap visible light spectrophotometer and some pDMAB as a colorimetric reagent on aqueous/alcoholic extracts.

Maybe do the alcohol extract + evaporation on 3 different batches of the same isolate to verify the method is consistent.

Reason: There's already way too many extraction threads in advanced, where they don't belong anyway. It's well known that cubes have a wide variation in potency, regardless of other factors. It's just the way it is. Nobody is going to test every batch. If in doubt, double the dose. RR
