If I'm using agar wedges to inoculate rye jars, do I still need to punch holes in the lid? Or is it alright to just loosely screw the caps with the center-piece upside down?

Yes, you will always need at least one small hole for gas exchange bro.
Try a 1/16 inch drill bit, or a really small Phillips head and a hammer.
Go smaller at first, then larger if you think it needs it.
If your hole is too big your sub will dry.

Four 1/16" to 1/8" holes are recommended for gas exchange. Screw the lid tight and use a filter over the holes to prevent contaminants entering. RR
