Agar isolation question
    #7910896 - 01/21/08 06:02 PM (16 years, 2 months ago)

So I'm going to use agar to do a strain isolation from a syringe; I've seen two approaches to isolation one in which a promising segment from the initial sample is excised and placed on the next dish and grown out, the process is then repeated until the isolation is complete. The second way I've seen is that several samples are taken from the initial plate and all placed on the next agar dish and so on until the isolation is complete.
My question is: is this just a procedural difference? Or is one method superior to another?

A single swipe of spores from a print, or a couple drops from a syringe might generate dozens or more sectors. I'd suggest transferring all of them to new dishes. You don't want to just isolate one strain, because it might be a dud. Isolate several promising looking strains and then grow each one out to determine the one(s) you want to keep for perpetuity in master culture slants.

So you suggest taking several sectors and transferring them to their OWN dish rather than placing multiple sectors in one dish? (I'm just clarifying b/c I've seen dishes with multiple sectors that looked like pieces of pie, like 6 sectors in one dish)

I'm totally stoked about the isolation, I love all the processes in my amateur mycology hobby, and I definitely like new shit. I'm not saying I don't wanna do it, I just wanna make sure I do it right.
