Dying Mycelium
    #7906410 - 01/20/08 04:47 PM (16 years, 12 days ago)

I have started 10 jars of rye seed from a spore syringe and have colonized all of them. However when i attempt to start 10 from each using grain to grain transfer most of the mycelium dies after a short while in the new jar.

did you use the same technique with all 100 jars you prepared? what are your incubation temps, jar size, etc?

Where did you get the spores from?

Sounds like either contamination in your jars or mycelial senescence.

well i have no contamination at all and im leaving the lids a little loose for air exchange. the only thing i can come up with is that the spores were causing this to begin with. ive been very sucessful with rye seed before.

It is strange that you're not seeing any growth. but if they colonized the first jars fine, i don't see why would have a problem unless it's senescence.

the spores are from a company in montreal
im using the large jars incubated at about 80F in complete darkness

that could be the problem
as fewer and fewer jars are colonizing
i suppose i would have to start all over again to solve this problem
i also tried taking a small sample of mycelium from a jar and growing it on agar
it does grow but it is very slow
it took about a week to make it to about the size of a dime

how much water are you using for 250ml s of rye
ive tried a lot of different amounts and it seems to make a big difference on the growth
