Multispore Vs Agar

I just wanted to show you guys what your cakes,casings,and grain basically look like when using a spore syringe. The spore syringes work well and are easy but once you guys start working with prints and agar things get better. You will most likely have faster colonization, bigger fruits, healthier mycelium.
here are 3 petris of Stropharia rugoso-annulata (non active). The one on the left is the master and spores were used to inoculate it. The one on the top was cut from the master about two weeks later. The one on the right was cut from the master about a week ago. The two that were cut are still sectoring and one is showing fatter rhizomorphs than the other. They are still far away from being true isolates. Notice how the master still hasnt reached the sides of the petri and look at the top one. The top one is two weeks younger but is growing faster than the master but the rhizomorphs are not as thick as the one on the bottom right. I will be keeping my eye on the fat rhizos on this one.
So basically the one on the left is what your cakes, casings look like when you use a spore syringe

isolating is wonderful.. I always feel like a mad scientist when I do agar cultures... Dr. Frankenstein to the front desk... ITS FROKENSTEEN!!!!

when you begin isolating do you want to take only the first spots of growth since that means they are the fastest colonizers, or would you want to wait and choose all of the heatlhy, rhizo growth sections?

wait for something to shoot out faster than the rest. Fast germinating isn't what you are really looking for. Once they start mating they will start growing rhizomorphic. Agar is neat because you will get sections that just look like contams. Just a white puff with no real shape. You can cut a chunk from that and rhizos will grow out everywhere.
You can kind of see where I cut the other 2 plates out of the master.
On the bottom plate which actually looks better than the top because it has fatter rhizos, you can see 2 sectors that look good. to the left of center and the bottom.
The top plate is still sectoring a bit and it is growing fast but the rhizos are thin.
I might cut from the bottom right plate and transfer it to another plate but I will most likely just cut the left and bottom sectors and put them to agar. Sure they will sector a bit more and could be isolated more but these sra's will be grown outdoors so I still want a good amount of genetic material to insure that they fruit abundantly. Sure some will be huge, some small, some will fruit early and some later but I am still guaranteed 100% success. If I kept isolating sectors and placed them out doors for 3-4 months it could all be a waste of time if it doesn't fruit in the environment that I provide it
