colonization?

i few weeks back me and a friend purchased a Starter kitfrom mushbox.com along with Cambodian 10cc Syringefrom spores101.com
The Substrate jars came pre-sterilized, and with 4 holes on each jar for the syringe. We injected around 1cc+ into 7 of the jars(all that would fit in the incubator, without stacking) We have it so the temp in the incubator is around 80F and is covered with a blanket to keep in the heat. now we have had the 7 jars incubating for more than a week and their is still no signs of colonization. does anyone have any idea what the problem might be? i think it could be the humidity, but this is are first time. also the temp outside the incubator ranges widely from 40 degrees to 75 degrees, because my friends house is heated by wood.

unless you have a kick ass heater in that thing the temp is not staying at 80 whne its 40 inside. just wait longer.

if we were to try again after waiting another week or so what temp would you recommend we start the incubation with?

i always incubate my cakes, jars at 28oC ive been told that the optimum is 30oC but i never incubate at that temp.
its better to be slightly under than over and risk destroying the mycellium.
another thought, maybe the syringe you got was a dud one. Try spotting the syringe on agar to see if its viable. thats what i do with my own stock (ive only had one strain)

also the core temp of substrate is 1-2oC higher since the cells a respiring and hence producing heat.

If you have to use an incubator, no higher than 81F. Make sure it is maintaing that temperature. Use a digi gauge that can take max and min readings.
You dont want massive temp swings as this can cause excessive gas exchange that may prove too much for your verm filter.
You dont have to incubate, room temp is just fine (75F).
There are other factors that could be your problem.
Sterile procedures - Sterilisaton method and time, glovebox, flame needle and alcohol swab before each jar.
Substrate moisture content, far to dry or far too wet (which can result in wet spot)
You may have bacteria in your substrate?? do they smell bad (be very carefull, nose full of bacteria is not nice!!!)
I agree it can take more than a week sometimes, generaly though i have always seen growth in 4-5 days.

For those who dont know what respire means.
Im not sure if that quite relates to how myc produces heat.
