Agar for Woodlovers
    #7866690 - 01/11/08 08:25 PM (16 years, 21 days ago)

I am currently attempting to cultivate the weraroa novazelandia, I can see that the spores germinated out a little bit in the syringe, but it just doesnt seem to want to germinate out on MEA, I made 6 petries 8 days ago and nothing on these ones but the other strains are doing great and I was wondering 2 things:
a: Is there an agar solution that favors the woodlovers vs. Cubes?
b: Are some species just tempermental when it comes to agar

i dont think so im not a pro at agar or anything close but it seemed to work fine when i used cubes, and carmel caps on the same type of agar and they did fine

mea should work fine. It could be the fact that you are trying to go from syringe to agar. That is messy and not ideal.
When I grew some wood lovers, Stropharia rugoso-annulata that is, i would sometimes boil up some wood chips and then use the water left over to mix with the agar. I have also used shreaded alder chips mixed with the agar and that work great for growth but it is tough to cut and isolate sectors but that isnt a big deal if you are putting them outside.
So mea should work fine. Give it a few days longer before you start to think things are bad. Here is my agar tech for using those chips.

1.instead of petris use 1/2 pint wide mouth jars, pf jars
2.mix up the agar and fill each jar with 3/16-1/4th inch of agar.
3.add a teas-tbs of these so that they are slightly submerged. A rough is great but you do not want the chips just hanging out above the agar
4. place the bands and lids on upside down. They can not seal. No holes, no tyvek, no polyfill in the lid and NOT AS A PRECAUTION
these things harbor contams also.
5.cover with foil and pc for 30-45 mins 15 psi.
6.let cool in the pc
7.work with these just as you would plates. place one finger on the lid holding the jar still while unscrewing the band with the other hand. Leave lid on but band off and work quickly. Replace band without knocking the lid around.

The main thing that sucks about this is the fact that the lib is not clear so you must remove it to get a good look at the sectors.
Once a jar is fully covered with mycelium you can add chips to the jar and the ycelium will reach upwards and keep growing. You can expand using these chips so grain is not needed.

Here are the chips

Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

I would suggest trying "cardboard agar", which is simply cutting a circle of cardboard and using that instead of agar.

http://www.shroomery.org/9393/FastFreds-Media-Cookbook
Quote:

Grey Cardboard (instead of agar)

Here are the detailed steps for making cardboard plates. Note that you can also use small jars in place of Petri dishes.

1) Measure about 100 mls. of tap water into a small jar.
2) For nutrients, , measure another 100 mls. of tap water into a second jar and add one drop of ordinary soy sauce to the water, and a quarter teaspoon (1.25 mls.) of molasses or light malt powder.
3) Find some gray cardboard, the thicker the better, preferably gray on both sides. Trace a Petri plate onto the cardboard with a pencil and cut out several disks to fit into your plates.
4) Weigh one of your disks and record the weight. Multiply this weight by a factor of 1.3 as a rough guide (you may need to experiment with the amounts here), and add the resulting weight of tap water or nutrient solution to each disk in its Petri plate. (Remember, 1 ounce of water equals 28.35 grams; one gram equals one milliliter.) Example: Suppose my disks weighed 0.17 ounces each. Multiplying 0.17 by 1.3, I get 0.22 ounces. There are 28.35 grams in an ounce, so 0.22 ounces x 28.35 equals 6.3 grams. That means I'll add 6.3 milliliters of solution to each disk.
5) Close up the disks in the plates, and let the water or nutrient solution soak in.
6) Pressure-sterilize the jar of plain water, and the Petri plates with moistened newsprint disks inside, for 10 minutes at 15 psi (allowing the cooker to equilibrate steam for 10 minutes before putting on the pressure regulator).
7) Cool the cooker, and remove the plates and jar of plain water.
8) When the water has cooled, add 3.3 mls. 3% peroxide to the jar, using a pasteurized pipette, to give you a final concentration of about 0.1% peroxide in sterile water.
9) Add about one third of the initial weight of the cardboard as 0.1% peroxide to each disk. Let the solution soak completely into the disks. They are now ready to use. [23]

I can't personally vouch for the technique, and I wouldn't think of molasses or soy sauce as that great of additives. It was simply included more or less as the source describes.

If that doesn't work there are plenty of media formulations to try. DFA might be a good bet since there is a lot of corn and rice in dog food which might provide the cellulose or other plant sources you need.

I also think it couldn't hurt to add some source of cellulose to any of the mixes.

P.S. The above text isn't the clearest, but just a reminder: you can't autoclave plastic petri dishes. So use glass dishes or sterilize the cardboard first and then put into sterile plastic dishes.

8 days really isn't that long. The already germinated mycelium that's in the syringe should grow out on the MEA. Just wait a little longer.

You could always do a couple of PDA plates as well to see if that species prefers it better. It's good to have a few diverse flavors of agar handy for times like this.

this tek works great for wood lovers and have had success with it the tek calls for live tissue but i have used lc colonized grain and have read that people have used multi spore also with success.
