LC to Agar to assess contamination?

Salutations all-

After preparing a few liquid cultures, I began to consider the volume of contaminated LC's cultivators experience, and present the following question:

Granted that taking one culture to agar is creating a risk of contamination, does anyone take Lc's to agar in order to assess their viability before taking it to grain?

I just make up 1 pf tek jar as a test, if it works out then i use the rest of the syringe.

So far i havnt had a contaminated LC. Though i have had 1 LC look ok but not grow in a substrate.

The agar however could be a good thing.If u did get contamination and myc grew aswell, you still have the option of cleaning up by transfering.

If you have agar skills, I wouldn't even bother with LC. An agar plate will darn near fully grow out from spores before you even see a speck of white in an LC. In addition, you can isolate for rhizomorphic growth on agar.

unfortunately, I haven't touched an agar plate in 7 years, but I suppose shaking the rust off old tricks is worth it.

my new gig is isolationg rhizomorphic mycelium on a plate and putting it to agar. To test the lc I use a pf jar or grain jar. I do not shake the grain jar and i only inoculate one side of the grain jar. I watch it for a week and if it passes the test I shake it. then I watch it. If it has and mold it will blow up once shook. Finnaly I smell it once fully colonized. it takes a few weeks to test for sure but it is worth it.

Stop experimenting half way through your first grow. Grow it to maturity, watch it, learn from it. Do this a few times then experiment with different ideas and figure out what works best for you.

The best way to to go about propagating huge amounts of spawn would be to start off from spores to agar, isolate out the best growth over 4 or 5 generations and then slice it into small squares (wedges) and add to a jar of rye, then use that rye with the isolated mycelium to spawn as many jars as it can....

Growing out optimum growth takes abit of time but in the long run you know your going to get the best colonization times and fruits, suffer now and laugh later, LC's are way to easy to contam in my opinion and you can never be sure. but all this is just my two cents
