I have heard a lot of sucecss with this, and alot of failure. Getting a clean sample is very important.

Larger gauge needle ..... and I have heard that you can 'flex' the stem and take the biopsy diagonally and this helps you to get a nice core sample.
Check out these large, 16 gauge, 'core sampling' needles found at the local feed store .... $ .50 ... price was right.

Uncle Rico, thanx to you also. I'm gonna head to the ag store right now to pick up some of those needles. Looks like that needle would be good for drawing up LC too. Hey shroober, good looking out, you are exactly correct.

In front of a laminar flow hood, or inside a glovebox, simply shave a small piece of tissue from the inside of the stem and place on agar. Within 24 to 48 hours, you'll have a culture. If there's any contaminants, you'll easily see them on the petri dish, and can transfer healthy mycelium away from it to a fresh dish. When the culture is pure, transfer to grains. RR

Yes MAN ....
Long needles are good for sucking up LC etc.....
A couple of things I noticed:
My LC jar had holes that were sealed with silicone .... original hole was too small. So ..... increase the diameter of the holes ... obvious
When inoculating WBS with LC through a polyfill stuffed larger hole (polyfill was stuffed pretty tightly), the polyfill was pushed into the jar by the larger gauge needle. So don't pack the polyfill as tightly or have another method of gas exchange in your spawn jar lids.
Don't be retarded like me .... make accommodations for the larger diameter needle.

RR, I do have some lab malt agar petris. Good?---But I do want to make a LC. Can I transfer agar to LC?
