And 2/3 of the Jars were Smited!
    #7822843 - 01/02/08 07:51 AM (16 years, 30 days ago)

I birthed 4 fully colonized cakes today, the other 8
contaminated with cobweb and green mold. Separated
each contaminated jar from the group each time I
spotted a contaminated jar.

I did all of my inoculation procedures inside of a
glove box. I misted the outside of the jars and the
incubator daily with bleach water.

The jars were ordered from MushBox and the Syringe
from Spores 101.

I did everything possible to maintain absolute
sterility, including HEPA filtration.

Is it possible that the problem is not on my end?

What kind of hepa filtration? If you combined a hepa filter with a glovebox, that is very likely the problem. Turbulence is a far greater concern than filtration.

If the syringe was contaminated, all the jars would have failed instead of most of them. Another likely scenario is that the verm barrier on several of the jars shifted during transit.

Lately, I've been making a lot of pf type cakes without the verm barrier. Simply cover the inoculation holes with micropore tape, and then after inoculation, put a second piece of tape over the first. This even lets you fill the jars a bit more with brf.

I do have a hepa filter attached to my glove box.

Well that might be your problem, a glove box is not sterile, but provides an enviroment in which the air is not moving, so contaminants arent floating around and landing on everything.
