Tips for Germinatiing Five year Old Prints???
    #7819111 - 01/01/08 05:58 AM (16 years, 30 days ago)

I have a few prints that are well over 5 years old. I have put them to agar and after 3 weeks they still havent germinated. I am assuming that it is becasue the spores we very dy and needed to be rehydrated.
I am thinking of mixinf some spores with a few drops of water and allowing the water to evapporate after a few days and then trying again.
You guys have any other tips?

If the spores are on the agar still, you can swipe some sterilized water over the top of them. I don't think more than one spore in a million will germinate after that long, so you'll have to play the numbers and use lots of re-hydrated spores. I've had limited luck adding activated carbon to the agar when working with really old spores. I used a teaspoon of powdered carbon in 1/2 liter of agar mix, and it seemed to help a bit, and certainly didn't hurt.

What is the idea/reasoning behind the addition of the activated carbon?

The general theory is that the charcoal absorbs certain inhibitory compounds, thus helping germination.

Seeds and spores often have certain compounds in them that prevent them from germinating until conditions are right, such as being washed away or diluted by water. By absorbing those inhibitory compounds they are thus encouraged to germinate.
