please help, brown slime like growth on agar plates
    #7808158 - 12/28/07 06:05 PM (16 years, 1 month ago)

I have some experience in growing shrooms from years ago and of course I have had contaminations but this is something I've not seen before. The agar plates get a brown slime looking growth around the inside perimeter and colonies growing the same type of brown slimy growth throughout the surface of the agar. This growth occurs after about two to three days after pouring the plates and will also occur on plates I leave uninoculated so I know its not from any transfer of spores or mycelium. The brown slime will run over the entire surface of the agar. Would anyone have any ideas on what it is and how I can avoid this? I do not see any other types of contaminations other than this brown slimy growth. Thanks for any help. If I knew how I could include a picture so any advice on that is greatly appreciated.

Bacteria.

Provided you are sterilizing the agar properly, it must be getting in when you pour the dishes. The most common cause is your own breath. You also need to make sure your agar bottle is filtered so it won't draw in contaminants as the agar cools.

Don't forget to wrap the dishes with parafilm.

Thank you for the reply. It's been quite a few years since my last growing and the first time in this apt. For the agar I use a small mouth bottle and plug it with a few cotton balls then cover it with aluminum foil. I sterilize the agar for 30 to 40 minutes, then let it cool enough to handle the bottle, about one hour. I do all my work in a homemade glovebox made out of a used cardboard box. I've been thinking of just buying a new box from U-Haul which is what I've done in the past but I do spray with Lysol before, during, and after working in the box and I leave the plates in the box. I've never had to use parafilm before but I do have some available and I will use it on a test set of plates that I will make today just to see if that helps. And, I have been doing my work in the bathroom after spraying the counter top and air with Lysol, its the only room I have that is small and contained. I will begin doing the work on the cleaned kitchen table in case the bathroom is so contaminated that it gets in the glovebox before I'm done working. Thanks for the reminder of using parafilm.

The ambient trichoderma and penicillium spore load is so great in my area that any petri dish not wrapped with parafilm would have contaminants growing within 48 hours of pouring. People in desert climates have better chances, but it always pays to take precautions. In addition to parafilm, I also slip the dishes back into the plastic sleeve they come from the manufacturer in. This gives double protection.

Before I try anything again I will be putting together a new glove box using a new cardboard box. It is about 48 hours that growth shows up and its usually around the perimeter of the agar so things are getting in and falling down and start growing. I plan on going at it again in the next couple of days, I will let you know how things go. Thanks for the help.

To follow up on this:

I tried many different angles including the parafilm but always the same problem with the same contaminant, a brown wet growth. I have never before seen such problems though its been awhile since I've been cultivating. In desperation I used MEA agar with a antibiotic and things are going well, no more of that bacteria growing on the plates. I've found the drinking water in this town is terrible (I've always only used Di Water for the plates) so there might be other problems, I plan on moving soon to another town anyway but this is just something I've never experienced before.

You can't make a glove box out of a cardboard box because you can't see through it....

It is also not fully sealed. But mostly, you'll be working in the dark if you try to make a glove box using cardboard.

yes, but cut off the top and cover it with a sheet of plexiglass. This allows a easily portable low cost replaceable box. Cut out access holes in the front and drape thin plastic over them that secures around the front when work is done, it ain't pretty but its functional.
