The day of my syringes getting here is getting closer and I want to make sure I get this right. Does the amount of cc's per hole if Im using the PF tek have anything to do with how long it takes to colonize?

The problem with adding more than the recommended syringe content to a jar is the extra water content. You don't want your substrate getting too wet! Why not follow one of the PF teks, measurements and all, and leave it at that? Don't try to reinvent the wheel ya know? After you get a few grows under your belt, experiment away!

Right. This has been covered a few times these last few days, so I'm going to be brief, and if you want more info, do a quick search.
One would think that the more spores you inject the faster it would colonize, BUT, adding too many spores will first of all mess about with the water content of your substrate (too much water will slow down colonization) and having too many spores also creates lots of different substrains (as every two spores will mate creating different substrates). Some of those substrains can be incompatible and instead of merging will compete with each other instead, reducing yield.
The only ways to speed colonization time with injection is making sure they spread well on inoculation. You do that by making sure the solution dribbles down the side of the glass (by injecting against the glass) so instead of growth starting on one little circle it will grow along the whole distance the dribble travelled.
In theory you could also speed it up by having more inoculation holes (say 8 instead of 4 as that would spread the spores out more) but you would have to remember to reduce the amount of spores per hole. You want 1cc per jar.

Thanks Nibin. I'll try 8 holes with a few jars. I have 40cc's coming for my first grow. Since it seems like you're pretty educated on this, would you recommend layering and casing in a big chamber(40 gal tank) to get a bigger yield or would it not make a difference?

With PF cakes? Unless you are adding a bulk substrate it isn't worth the hasstle.
