agar?!?! First time

I just made a agar solution yesterday with 1L of water 22g of agar and 20g of Brown rice flour. Is the agar supposed to look really solidfied like clear jello?. I injected 3cc of syringe based panama mushroom spores. ? is will it possibly grow? keepin the agar in 80 degrees. The reason why i ask is the spores are kinda floating on top of the agar is this normal?

You will probably get some pins, cubes pin on agar very easily.

Wait just reread your question, yes your agar is fine. It should be jelly and the mycelium only grows onthe surface of the agar.

syringes are not the best for agar. Everything will germintate on it. If the syringe is slightly dirty then you are fucked.
Where did you get the syringe?

3cc is way too much for agar, you just need a drop of syringe. Problem with syringes on agar also is that the water will swirl around and disperse any contams.

Did you use a petri?

I wouldnt say its fucked.

A contamnination isnt so much of a problem, since you can transfer a small peice of tissue to another plate, grow it out. If contam shows up again, do another transfer, until you have a clean sample.

With multispore you will probably want to do quite a few transfers to try and isolate it a little.

With that much spores, though it will make it quite difficult.

Idealy u want very little spores so that you can isolate the different substrains out in just a few transfers then use each isolate tested in say just 1 small casing or even pf tek to check for traits that may be desired.

Keep at it, look up the agar teks and other peoples post. Theres a wealth of information on the subject.

this is a easy petri dish. I was out and I needed to get shiitake cultures going. This is also done with a syringe, about .01cc, just a drop. Notice how it is goring on the sides. This is not ideal, the culture rolled to the edge. The agar level is also too thick, no more then 3/16" when you pour them.
This tech is easy thought. make your agar but do not cook, pour it into wide mouth 1/2 pints, put the metal lid on upside down/seal side up. You do not want them to seal!!! Put the metal band on and cover with foil. pc the whole jar all 12 or how ever many for 30 mins.
Let cool inside the pc. Work with them in a glove box, hold the metal lid on while removing the band, one finger in the middle pushing on the lid and the other hand unscrewing it. always leave the lid on except when qucikly inoculating it. Never fully remove lid, just tilt up slightly.
They work okay but the only draw back is that you can not see through the culture or the top unless you open it. Opening is bad unless you are clean. A petri you can see through the top and see what is going on at all times.

I think I'll be useing that instead of ordering prepored dishes. I don't have a flow hood yet and don't want to waste time on contam. Agar.

here is my glove box. I do not use gloves or a front. I just put the plexi cover on it and load it up and one spary of alcohol. If i experience a contam problem then I will build a front but so far so good (2months)I keep my burner outside the box. The kitchen trash can is right next to my work area. I find that it is a little too long and too short in height in the working area. I am going to shorten it by 6 inchs which will steepin the angle and give me more room above. My culturing tools hit the plexi sometimes

I only work with oyster, shiitake, lions mane, portobellos. I am sure this set up will work for all species

It that made of wood?
If so how did u seal it?
I've been using a rubber made tub and don't really like it.

i used to use a rubbmade but it is iffy. it is made out of partical board but its coated from the factory. It is called melamine. It comes in all sorts of colors, designs. Homedepot sells it as should any good building supply company. It is the same shit that is found in your kitchen cupboards if it was made in the past ten years. it is about 30$ for a 4x8 sheet. More then enough to make two boxes. I just predrilled the screw holes and sealed the seals with silicon.
it is easy to wipe down.

If you are too cheap to buy this then you can use particale board with a few cots of glos/semi gloss latex paint. But the pain+wood will be the same as just the wood.
make sure your highest point is high enough for a grain to grain transfer, so a 12 inch tall jar will need at least 6-8 inches of head room to allow you to pour from one to the other.
My opening is about 6 inches. I might make it a tad shorter in the front. This is my plan for it. I will basically cut the red line. This way i can reach to the back and it will be/seem higher.

Please note, my house is slightly cleaner then average. Not a clean freak but I have seen some houses. The open front is not recomended but works well for me since I do not live in a pigs sty. i also do not use gloves and I have hairy arms. I just spray them down with alcohol before and 1-2 times during use

Thanks, I'll be making somthing like that soon. I use to use melamine for reptile cages. Didn't think of it when I was making my 1st one.
