Colonization Help
    #7732350 - 12/08/07 09:18 AM (16 years, 1 month ago)

Hey everyone, my names Pete, i've been browsing through this great site for about a week now and decided to create an account,it's been very useful for me. Anyway, I am having some problems with colonization. It has been 6 days since I inoculated my jar and I am not seeing any mycelium growth at all. I am not sure what I am doing wrong, but maybe if I list my steps and materials, somebody can help me out. First off, I used organic precooked brown rice w/ no preservatives, it is the kind that comes in those little bags that you boil. I made sure to blend it to a powdery form in a blender. The vermiculite I purchased at Lowes, it was by Sta-Green I believe. I didn't have a pressure cooker, so I had to sterilize the jars in a pot using the steam method. I made sure to wait 24 hours before I inoculated the jars so the heat wouldn't kill the spores. The spores I purchased from spores101, and it is the Cambodian strain. I injected a little over 1.5 cc into the jar. I put it in a dark cabinet and check on it twice everyday or so, but I am not seeing anything. Can someone please help me out? I am a first time grower, so I have no idea what I might have done wrong. Thanks very much!

Give it a few more days. I've had jars take up to 2 weeks to show the first signs of mycelium.

Did you inoculate against the glass as per the PF Tek?

Yes Premedman1 I did, but if I may ask, what would be the difference in doing it otherwise? I sterilized the cabinet with Lysol just as a precaution, could this have affected the spores in any way? Also, should I let the jar receive some air every now and then?

If you inoculated the center of the cake, it could already be colonizing and is just not visible yet.

The reason to inoculate against the jar is you can see when mycelium developes, and you can spot contaminates sooner.

What do you mean by "let the jar have some air"? You didn't cover your 4 holes in the lid with anything, did you?

Spores can take a while to germinate sometimes.

In ideal conditions, you will signs 3-4 days at the earliest.

Give it another week.

With steam sterilisation cook for 1.5 hours to make sure it was properly sterilised.

It wont make much difference sterilising your incubating cabinet, the ry verm layer acts as a filter, if any contaminent spores get in the jar, they will be stopped by the verm layer.

No, I only covered the lid with aluminum foil during sterilization and have removed it since. By having some air, I meant opening the cabinet where it is being kept for a few minutes, not the lid of the jar.

Thats absolutly fine. The foil is only really needed during sterilising to stop moisture getting into the jar.

Yes, I let it steam for about 1.5 hours, maybe two at the most. I made sure to let it cool for a day before I injected the spores. Do you mean 3-4 days from the time I inoculated, or 3-4 days from now? All of the guides I have read so far said to use organic brown rice with no preservatives, would it make a difference if the box said the rice was precooked? The reason I said if the Lysol might have affected the spores is because since it is supposed to kill germs and bacteria, the spores are bacteria, I guess?

i was in the same situation a couple weeks ago, hadn't seen any growth and was starting to get worried. at about 2 weeks after innoc, the jars exploded with growth, just give it some time and you'll be happy
