Joosus' PF Tek.. PART 1.. substrate prep and inoculation...

First off, a word about sterility. KEEP EVERYTHING CLEAN!!! That's it. Clean all work surfaces with alcohol or some type of disinfectant. Clean all tools with alcohol. Turn off all fans and air conditioners 10 minutes before you start working on anything. This will help the air around your work area settle down. Follow common sense when doing this and you will succeed. 90% of first timers FAIL because of non-sterile techniques. Wash your hands all the way up to the elbow just like doctor shows and wear a hat. I got some crazy hair, so I wear a hat to keep it tied back while I work. make sure all bowls and measuring cups are clean as well. A HEPA filter (can be purchased from Target, Walgreens, Home Depot, Etc..) can also be used to catch spores and other whatnot over by your work area. Or you can make/use a flow hood. Search shroomery.org to find the FAQ on how to make that.

THIS RECIPE IS FOR 6-1/2 PINT CANNING JARS. FOR MORE, JUST DOUBLE THE RECIPE!

On to the good stuff!!!

What you need:
6 wide mouth 1/2 pint canning jars with a 2 piece lid


A coffee grinder (optional unless you cannot find brown rice FLOUR)


A bag or 2 of organic LONG GRAIN brown rice


A bag of vermiculite


Coffee filters
*** We all know what these look like, right?

A big mixing bowl


A fork
You must have one of these in your life...

A bottle of rubbing alcohol



Big steamer pot or pressure cooker (pressure cooker preferred)



Optional stuff to help the process:
Heating pad


rooting mat thermostat


And of course, a source of fungi. Spore print, spore syringe, live culture, etc.



I will be doing a grain-to-grain transfer of the mycelium from one jar that I had to the new ones. You can also use a spore syringe in this method, but other precautions must be taken, that will be explained later on.

First, CLEAN THE WHOLE WORK AREA!!! Spray lysol in the air surrounding the CLEAN work area, and wait about 10 minutes for everything to settle. Wash up and get everything in the work area.

Next, in the big mixing bowl, add 3.5 cups of vermiculite to the bowl. (you can double this for 3-1 pint jars or 12-1/2 pint jars)



Now, start slowly adding water and mixing until the vermiculite cannot hold anymore water. You should see it start to puddle up in the bowl. Keep mixing until NO MORE water can be soaked up, then strain out the rest of the water.


I just tilt the bowl sideways and use a fork as a little filter. You want to maybe see JUST A LIL BIT of standing water, but just a couple of drops of standing water is where you want it.



Next, take your brown rice and put it in the coffee grinder. We do this to turn brown rice into brown rice flour. grind it as fine as you can.



Measure out 1 cup of brown rice flour and put it into the mixing bowl with the now wet vermiculite.


Stir until the vermiculite is evenly coated with brown rice flour.


Now we prepare the jars.

Take the big flat piece of the 2 piece lid and drill a 1/4" to 1/3" hole in the center of the lid. This will help the jars breath. Do this to all the lids.


Now, fill all of the jars up to about a 1/4" to 1/2" from the top. DO NOT PRESS IT DOWN! Spoon it in very easily, as you want the mixture in the jars to be light a fluffy.


Cover the jars with the lids that you just put the holes through.


Take your coffee filters and rip them in half.


Set one half per jar on top of the jars, making sure to cover the whole lid with the filter. Be sure that the filter covers the whole lid, so that it has a good seal.


After you have the filters on the jars, now put the 2nd part of the lid on, so that it secures the coffee filter. You should NOT be able to touch the part of the lid that you drilled through without removing the filter. A complete seal.




Here is where controversy comes in. I have done this for years, and NEVER used a pressure cooker, and have had great results. Although I HAVE used a pressure cooker for any type of grain I have grown on (rye wheat, bird seed, etc.). A pressure cooker is pretty much a necessity for grains, but this method for brown rice flour works great without one.

Get you big steamer pot and fill with water to just below the rack that you put the food on (in this care, jars). Bring just to a boil.


Put your jars in the pot so they are not touching each other and bring the boil down to a simmering steam (it may take some time to find this. You want it to be almost boiling and steaming alot). Now cover the pot with the lid. Leave them in the to steam for 45 min to 1 hour. (again, its your choice. Just no less than 45 minutes. BUT IT HAS TO BE STEAMING ALOT!)

I want to stress the importance of the heat. ALMOST A BOIL, BUT STEAMING ALOT. That's perfect. We are trying to kill any spores or bacteria that is in the substrate and jars so that when we inoculate, we will have a nice, sterile substrate to grown on. This makes us happy.

You would do the same things in this method with a pressure cooker, but cook the jars at 15 psi for 45 minutes to an hour. When the pressure cooker is 100% cool, then remove jars and proceed to inoculation.

After 45 mins. or 1 hour turn off the heat and let the jars cool over night or until cool. LEAVE IN THE STEAMER. Take out when completely cool.

Re-clean you work area. Sterilize, wash, etc..

Now here is the tricky part. You should wear a dust mask over your face or at least your shirt, either hold your breath or breath VVEEERRRRYYYY slowly while any of the jars are open.

Get you live culture (or whatever you are using to inoculate with) and set it next to your jars in you work area. CLEAN CLEAN CLEAN WORK AREA!! THIS IS VERY CRITICAL.

Sterilize your fork (or tweezers) with rubbing alcohol by rubbing it all over with a paper towel or coffee filter. Set it down on a clean coffee filter or paper towel.

First, take a jar and set it next to your live culture.

Take the 2nd part of the lid (the ring) off and set next to the jar.

VERY CAREFULLY open the second part of the lid. Not all the way. Just break the seal. DO NOT TAKE LID OFF!! DO NOT TAKE FILTER OFF!! OPEN THE LID AND FILTER AS ONE PIECE!!

Now, take your fork (or tweezers) and pull a chunk of mycelium out of the culture.


Now VERY SLOWLY, open the lid to your sterile jar and drop the fungi on the fork in the jar. YOU DO NOT NEED TO OPEN THE LID ALL THE WAY! JUST ENOUGH TO GET THE FUNGI IN!




Put the lid back together just like it was and set aside. Repeat for all jars. PRACTICE STERILE TECHNIQUES. PLAN EVERY MOVEMENT! THE SUCCESSFUL CULTIVATOR IS THE METHODICAL ONE.

Next, prepare a dark place for the jars to go. They need to be kept in pitch black at room temp (70-80) for a while, until the jars are completey filled with white mycelium.

This next step is just something I have always used. It makes them grow faster.

In your cupboard, box, grow room, etc.; lay the heating pad down. Connect the heating pad to the thermostat and set for 78 degrees. The jars tend to put off a few degrees of heat on there own, so setting it a little lower than 80 is about right. For me, anyway.

Set the jars on the heating pad, and put the probe for the thermostat in the middle of the heating pad. Set it to 78 and set the heating pad on low heat.



Let them sit there for about 2-3 days, and you should start seeing growth (only if you used a live culture or liquid/agar culture. If you inoculated from spore, then allow about 5-10 days to start seeing any growth at all.)

After about a month, give or take a week or 2, you should have some fully colonized jars. Like this:

When you have 1 half pint jar, and let's say you break up all the mycelium.  How big of a jar can you upgrade to, or recommend upgrading to, and how many do you recommend spawning from one half pint?

Thanks!  Awesome Tek, glad someone has done this.  Also have you or anyone tried going from PF Tek and doing the same method but over to WBS?
