Is a glove box absolutely needed? No. If you can afford Lysol, you can make a glove box! Is a glove box recommended? Hell yea! You can inoculate and take a chance, but the first time you fail to contamination, you will kick yourself for not taking all the precautions. Glove box being one of them.

I personally haven't had a problem with the bathroom technique for simple nocc ups and lc using. I have a glove box and am still getting used to using it for when i step up to agar.
but if i'm doing anything more than 12 jars, its kind of a pain
I guess its a toss up between safety and comfort, and comfort usually wins for me.
remember, flame is your friend, but not around alcohol or aerosol

I open air noc all time using alcohol sanitizer gel. I flame sterilize, then wipe the gel on to the needle which immediatly cools the needle, and coats it in sanitizer. Then right into the injection port it goes. Just don't get lazy.... Do this before every jar. Haven't contamed a jar in a very long time.

For innoculations, no, you don't need a glove box. I would recommend making one. I will post a picture of my first ghetto glove box. I made it from a box and two garbage bags, one glued into the box to provide a surface that is easier to clean and another clear one as the lid. It works well and I haven't lost a G2G transfer since implementing the use of a glove box, even that ghetto one I made.

I would never consider inoculating anything without a flowhood or glovebox. However, that's just because I'm very intolerant of failure. Your success rate will be higher by following proper procedure, and a glovebox can be made for ten bucks, so why waste time without one? RR
