You have a better success rate transferring a wedge to a LC then inoculate grain to spawn to hpoo or put a wedge in a qt of grain and then G2G then spawn to hpoo.

Whatever you transfer agar to should be sterile. PC the Hpoo in a jar, transfer, when you get good colonization transfer to your pasteurized Hpoo.

I wouldn't transfer to poo.  Remember, we spawn to a bulk substrate in terms of ratios..... 1:4 being the norm and anything above that being brave when considering the longer colonization times.  A wedge of agar to Poo would be like a ratio of 1:100000.  Kind of crazy when you think about it that way don't you think.

As said, if the manure wasn't sterile, it would contaminate the back side of the agar wedge with bacteria before the mushroom mycelium could crawl off it and get a foothold into the manure. Use agar to inoculate grains, and then use the colonized grains to inoculate pasteurized bulk substrates.
