First time grower with a question

First, just wanted to say what an amazing site this is. I have read through almost every growing guide out there as well as some of the custom ones posted in some of the forum signatures.
I decided to go with the basic PF Tek with one minor modification. Instead of using tape to cover the holes in the lid, I used the guide to the self healing lid with silicone. Anyways, my question was this. I knocked up the 24 jars I'm trying to cultivate tonight and I noticed something I wasn't expecting (though I'm not sure why heh), the jars after sterilization had a really good vacuum seal. I was wondering if this would greatly hinder or halt any growth that the spores would have.

I would say YES! I am a mushroom noob too. I started a few months ago and the mycelium in the jars stopped growing. I decided to drill 4 1/2 holes in the lid and they are growing fast again( I have a vermiculite filter).The small holes caused a growth stunt.

Many people (myself included) do well with not covering the holes in the jar lids and just relying on the Verm Layer on top. This allows for maximum FAE inside the jars. Just let the jars sit on a book shelf or something while they're colonizing - works great! (clean the area BEFORE hand of course)
(Cover them with foil in the PC though!)

Thanks for the feedback, guess Ill let the jars go and see how it works out. If its a complete failure, I'll try using either the tape method or just leaving it open.

you should be ok.. the FAE is helpful, as it speeds up colonization, but if you have moisture content right and no other issues the jars should be ok, just a lot slower to complete

You do not want fresh air exchange (FAE) when colonizing. You want gas exchange. There is a difference.
Your jars are likely to stall without gas exchange.

Do not seal the inoc holes, leave them open. There needs to be sufficient gass exchange or as ziggen says, the myc will stall.
The dry verm layer acts as a filter stopping contaminents getting to the nutritious layer of substrate.
Micropore tape is fine as an extra precaution, but do not seal them complelty (micropore tape wil breathe)

Again, thanks for the input. In that case, would you guys suggest just carefully (and in a sterile environment) poke some holes in the silicone seal and cover with some micro-pore tape?

Poking any significantly sized hole in your self-healing injection point will only serve to ruin it and prevent it from being useful in the future. If you are going to futz with something, you may as well poke a few holes on the lid itself and cover with micropore tape. You may want to do this in a rudimentary glovebox to reduce the possibility of contamination.
Also, search for Tyvek and read some of the various ways people use Tyvek (usually from Tyvek mailing envelopes) to aid in gas exchange during colonization.

Just loosen the lids so that a bit of air gets in without dust getting in. The dry layer of verm will protect the cake.
The biggest problem with the vacuum is that unless you break it first, the spore solution will be sucked out of your syringe. Also air can be sucked in fast which might be an issue.
You need gas exchange or your cakes won't colonize.
