BRF paste, agar replacement attempt.

Ok so I am trying to replace agar with a formula of BRF, distilled water, karo, and crisco. I put this in a sideways pint canning jar, and fashioned a lid to fit a modified butter knife through to scrape out mycelium when and IF it grows. The hole will be plugged with polyfill, and taped until such time as a glove box is used for mycelium extraction.

My question is this. How best to add peroxide to this solution to make it possible to transfer live mycelium without the worry of germinating "nasty" spores.

I am pretty sure h202 has to be applied as the mixture is cooling so as not to overheat it correct?

Any comments, ideas, or poking fun would be appreciated.

I would omit the Crisco,
and the only thing I would use peroxide for is if I were trying to clone from a wild fruit.

ya, don't use peroxide.

Peroxide is usually only good for killing an attacking cobweb mold.

Not if your making peroxidated agar dishes. There is such a thing, along with antibacterial agar.

I'm no fan of peroxided agar. If you mix it strong enough to suppress bacteria, it also suppresses mushroom mycelium, and the whole point of agar work is to encourage the mushroom mycelium to grow as fast as possible so you can get it to outrun contaminants and obtain a clean culture. Gentmycin sulphate makes a great antibiotic because it's autoclavable.

I fail to understand the point of the brf/crisco/karo stuff, even if you could get something to grow on it. The purpose of agar is so the mycelium grows on the flat surface of the plate so that you can see the entire organism. BRF is going to support three dimensional growth, so you may as well use pf jars or whatever. Agar is easy to obtain and use.

The crisco makes it possible to spread the substance to a perfectly flat plane. I just wanted to find another way to isolate substrains using things many people have easy access to. I live in the middle of no where so it is months sometimes before I get to head in to "town" to get things like agar. In the mean time I like doing things like this just to see what happens. People seem intimidated by agar. Maybe its just me.

Well I sure as hell wouldn't call it a agar replacment. But I DID get something to grow on it. And no contams either! No 3d growth as far as I can tell. "Nothing showing on the bottom of the glass" The downside is that it got bumpy while being PC'ed. The jar that I had set up as decribed up top was swabbed with the smallest amount of solution, but never grew any mycelium. This one, a tester, was given a slightly larger amount of solution.

Question: I have never isolated substrains before, but to me this looks like only one. Does anyone else have an opinion on this? I will post more pics as it gets bigger.
