agar contaminatons
    #7636138 - 11/14/07 05:08 PM (16 years, 2 months ago)

I have a few agar plates going with a couple diffrent strains in them.

One i did of pesa with a multispore inoculation off a print, it was growing some fuzzy white where I put the spores. But then a patch of myc appeared on the side away from the inoculation point and was kinda orange, then like 2 days later the white myc grew over the orange patch and I am just left with a total white patch.

Does myc fight off and take over contams in the agar plates?

When this agar plate is done will it still contain the orange contam or has the white myc killed it off?

Can i take a totally pure white slice out of this plate when it is totally colonized and make another plate out of it, or will the contam follow?

Agar isn't used to expand mycelium, it's used to isolate what you're looking for from what you're not. What I do is immediately transfer each new growth I see to a new dish before it has a chance to mature. By doing this, some dishes will have nothing but contaminants, and as soon as they're discovered(green, brown, grey, etc.), the dish is discarded. Other dishes will have various strains/substrains from the print of spores you used.

Yeah I have used a TSA plate on all my syringes to see if they are contamed but the different strains look so different. I am gonna make up some nutrient agar plates and try those to see if I get a different result.
I know you already told me the TSA plates are for bacteria and not fungi.

Hopefully the guy at mushymart poured some 3 section plates tomarrow.

Thanks
