P. Mexicana - A Strain -- Need Help/Advice! (pics)
    #753915 - 07/17/02 02:35 PM (21 years, 8 months ago)

Hey guys.

A few jars of P. Mexicana A Strain from Sporeworks going here.

I've never encountered the mycelium of these so I would like some input form the pros on this.

I have a few pics of some Rye Grass Seed w/ Brown Rice jars going.

Some are a little blurry... I will get more asap from my friend.

Jar #1

This resembles some ps mexicana i've seen on here but im suspicious about it... have a look...
---

----

Jar #2

This one kinda looks like p. trops mycelium almost... however it's supposed to be ps mexicana... let me know

More pics later.

A third jar is filled with a water reservoir!!! Pics comin up.

Let me know what you think, thanks a lot

Looks good to me. You should be seeing sclerotia very soon.

Workman,

The mycelium in the second jar looks so different. Is it ps mexicana as well?

Thanks

Yes, it appears to also be mexicana. Mexicana mycelium is a bit variable in appearance depending on the substrate, moisture level, temperature, etc. Once you start seeing sclerotia forming you will be certain of the identification. How old are those jars and at what temperature?

I'm not sure of the exact date... but i estimate somewhere 40+ days

Do you know if it would be possible to transfer some of this mycelium to a honeywater tek jar or a 4% malt jar and cultivate the mycelium like that?

What about grain transfers, how does mexicana mycelium do with that?

Anything you can do with cubensis, you can do with mexicana. It just looks a bit different, grows a bit slower and isn't as contamination resistant when cased. 40 days is a bit long, but your substrate is very dense and its going to take awhile to colonize the bottom. I am suprised that you don't have at least a few small sclerotia forming at the top of the jars by now. What is the temperature?

Though I have never worked with this strain, it looks to me your jars are toast!!! The sclerotia I have seen forming in jars looks nothing like that. They look full of bacteria to me, but hey what do I know.....Workman seems to think they are fine.

Looks to me as though there's too much water acumulated at the bottom of the jars. The rye looks mushie thich and wet.
There was probably too much water to rye ratio. The bottom might never colonize.
I would transfer what's good now into another jar or case it now after soaking in some h2o2 for a little while.

try using less water to grain ratio next time. What works best is to pre-boil your rye berries and then pressure cook them in your ja(s) without adding anymore water.

He is apparently working with an Una style substrate of rice and grass seed. It is supposed to be thick and unshakable like that. What you seem to think is bacteria is an accumulation of metabolites that often occurs with this species. Those jars look fine as mexicana but if they were cubensis, I'd be afraid. Also there is no sclerotia in the pictures that I can see, just uncolonized substrate and maturing mycelium (its supposed to get brown like that).

As Workman said it, this is a Rye Grass Seed + Brown Rice jar... It is not your usual rye grain jar, that is something i mastered a long time ago
Contamination in rye grain is very easy to spot.

Workman,
Yeah the mix I am trying out is 3 parts rye grass seed 1 part brown rice. Temperature has been around 80-82F.

One note, after observing an abundant amount of water in Jar A, i took off the top metal lid of the jar, and it's now just a single filter disk with band.

What effect you think this would have? I figured that the jar needed some airing... It's been open about 2 weeks and some of the mycelium seemed to dry up a little all the way at the top.

Jar B (the whiter one) still has the top lid on upside down.

Let me know

RICE IS VERY STARCHY. You have compacted areas that are resistent to colonization. Put the lid back on the top jar, turn it upside down and give it a few thumps. Make sure there is a space between the bottom of the jar and the substrate block. Leave it upside down to continue colonizing.

It is very difficult to get even colonization in rice based substrates without liquid mycelial innoculation. It is hard enough to shake Rye grass seed in high volumes.

If starting from spore syringe, or agar and using jars, USE LESS SUBSTRATE. No more then half the jar full. With the bags you can move the large quantities around to distribute mycelium, JARS are very difficult to shake.

Just stick with them, I had one jar take three months to finish colonizing, and start forming sclerotia. Another jar innoculated at the same time, WAS FINISHED ALREADY.

I haven't attempted the rice/grass seed mixture in a jar so I don't really have any advice. Teonan seems to have some experience, listen to him. Drying is bad though and will reduce sclerotia formation.

The white lumps in the top image. They should grow and darken over time. At least it looks like it from here. Its a bit difficult to tell.
