Spawn Preparation
Materials: Rye Grain, WBS(Wild Bird Seed), Pressure Cooker, Strainer, Paper Towel, 1 Cup Measuring Cup, Synthetic Filter Disk or Tyvek Filter, Solo Cup, Empty Gallon Jug, Canning Jars, Aluminum Foil, water

Formula #1 - 100% Rye
Materials: Rye Feed or Rye Grain and a Pot of Water
I measure out 14 cups of rye grain. my pressure cooker can do 14 quart jars at a time so i add 1 cup to 1 jar. as long as it doesn't fill the jar more than 2/3.


Boil a pot of water. when the water is boiling, add the grain to the water to soak for 25-30 minutes and turn the flame down to low or medium.


Strain well for about 1 and a half hours. Don't get skimpy on this step or you will be wasteing your time with the whole project.


Formula #2 - 50/50 Rye/Wild Bird Seed
I like to mix rye and bird seed because it balances the water content out nicely in my opinion. you can do your own mixture or you can use just rye or just bird seed. For this tek i am going to mix them in a 50/50 ratio of bird seed to rye grain. I like to use bird seed that contains no sunflower seeds, which helps speed up the process a little bit and requires less work. Sunflower seeds tend to contaminate easily for that reason i do not buy bird seed that contain them. If you have sunflower seeds in your bag you can easily remove them once you drop them into water (they float to the top, just scoop them out). I like to use Wild Delight Outdoor Finch Food but any bird seed will do. Wild Delight contains Millet, Canary Grass Seed, and Niger Seed. I use 7 Cups of bird seed and 7 cups of rye grain and add both to a rapid boiling pot of water. Make sure the water covers the grains/birdseed completely. after all you don't want the water level to ever drop below the grains/birdseed level at any point during this process. Simmer for 20-30 minutes on a low flame. Once the grains are hydrated, strain for 1 and a half hours.

Formula #3 - Wild Bird Seed
To prepare wild bird seed the steps are simple. first boil a pot of water. when the water is at a slow boil, drop in your messured mount of birdseed. keep the flame on low and simmer the birdseed for 20-25 minutes. when 25 minutes is up, strain the seeds in a strainer for 1 and a half hours. Don't get skimpy on this step or you will be wasteing your time with the whole project.

for messuring birdseed i use more than I would if I was using rye grain because the size of the seeds is differant. 2 cups of bird seed per jar is sufficient but the general rule is to leave at least 1/3 of the jar empty. you want a lot of air in the jar so that when you break your grains apart for spawning there is room for the grain to move and break apart easily. If overfilling does occure yo will end up spending a bunch of time scraping the birdseed out with a sterile spoon.

Loading The Jars
When the hour is up i like to wrap my hand in a few paper towels and run my hand threw the grains to make sure they are very thouroughly drained of excess water. I do this every 5-10 minutes once I hit the hour mark of straining time. so all together you should get at least 3-6 passes made in the remaining 1/2 hour of straining time.


i like to use an empty gallon jug and cut it in half just below the handle to make somewhat of a funnel when flipped upside down. I use A plastic solo cup to scoop the grains out of the strainer and into the funnel. Simply distribute the grains evenly between the jars. I like to line them all up in a row so that i can see all the grains in the jar to see if they really are even. I think i might suffer from OCD so this isn't a must but even is good. If that is not possible for you then use the solo cup as a measureing cup. use 1 scoop per jar until the grain is gone. if you have done one scoop per jar and still have some grain left over, continue by starting back at jar one until the grains are gone.




Once the jars are all evenly filled, place a tyvek/synthetic filter disc over the mouth of the jar. Place the lid over the filter and screw on the band to hold it all in place. once you have all your jars prepared in this manner you can proceed to cutting tin foil squares. I like to pull out a 4"-5" piece off from the roll and fold it in half and then you can make two covers from the one piece of foil. Now place the lid of the jar in the center of the foil and wrap around. The point of this process is to prevent steam droppletts from dripping off from the lid of the pressure cooker and messing up the moisture content of the grains.

Place all the jars in the pressure cooker and sterilize for 90 minutes. If you are really pressed for time 75 minutes will do, but I always figure a few extra minutes can't hurt. Once you have reached 90 minutes, allow jars to cool down to room tempurature. Once cool to the touch, you can move onto innoculations.


inoculation
Materials: Spore Syringe, Alcohol, Paper Towel, Nexcare Micropore tape, and Jars To inoculate
So your jars are cool and you want something to grow, well this is how.

Quick inoculation
The exposed filter under the gass exchange holes in the lid are wiped clean with alcohol. The needle is wiped clean with alcohol from the tip of the syringe to the end of the needle. this is to prevent stabbing yourself and from pushing thing down the tip of the needle and into your sterile culture. 1-2 ML of liquid culture of spore suspention fluid is injected into the jar. Remove the needle and quickly place on a strip of micropore tape over the gas exchange hole for an additional filter and to plug the hole in the filter punched by the needle. lable the name of the strain and the date of inoculation on the tape for easy reading and easy cleaning.

Cautious Appoach
First make sure the jars are cool, if you inoculate a hot jar the projec will fail due to excessive tempurature. Grab some alcohol and a paper towel. Get the jars you want to inoculate and remove the tin foil from the tops as that is no longer required. Begin to unscrew the lid but hold the lid and filter down in place as you will inoculate right threw the filter. Once you have removed the band, lightly lift the lid without removeing the filter. now take your paper towel and alcohol and get a little dab of alcohol on the paper towel. Wipe the needle down starting from the syringe end to the tip of the needle. This helps prevent getting stabbed by the needle. Wipe it a few times and then stick it threw the filter disc so that the hole in the jar lid is not going to expose the hole in the filter disc. Inject 1-2 ML of spore suspention fluid into each jar and place the lid and band back on. Make sure to wipe the needle down before each inoculation for extra safety. Once the jars are inoculated and the bands are replaced back onto the jars, shake the well to mix the spores around inside the jar.

Incubation conditions
Place The jars in a semi-sterile room. Incubate the jars at 84-86 degrees farenheit until full colonization of the grains. If you can't incubate them, then just keep them warm until full colonization is reached. If your jars reach temperatures higher than the mid 80's, you start conditioning the jars for growing other fungi and mold. A lot of what we would call contaminate fungi are practically inactive at room temperature, but in higher temperatures (110F-130F) they grow better than mycelium. Mycelium will occur a thermal death at a temperature of 106F but, if your temperature is at 86F you won't have to worry about that. Usually this whole incubation process takes anywhere from 1-3 weeks. More than likely closer to the 3 weeks unless using a Liquid culture or a multi-spore inoculation.

There's a couple of points I see that bear mentioning.

Adding dry grains to boiling water is a very good way to get burst/ruptured kernels. The grains should be hydrated slowly to prevent bursting.

You don't want to puncture your filter disks. In addition, rotating the lid won't stop contaminants from going through the hole you make in the filter. Air, and the contaminants within will take the path of least resistance, which is through the hole rather than through the filter material. It also helps to have the filter disk on the outside of the lid so it stays totally dry. A filter on the inside runs a risk of getting damp from condensation, which then allows contaminant molds and bacteria to colonize through the filter material and into the grains below.

Actually, thermal death comes much higher, despite what you'll read elsewhere. I've heated mycelium to 120F for half an hour in LC to kill off virus. That temperature kills the virus but leaves the mushroom mycelium unharmed. However, 86F is a few degrees past the point of fastest growth with cubensis, and it doesn't account for the 2 to 5 degrees of temperature rise due to thermogenesis. Growth with cubensis begins to decline at 83F, and that's when measured inside the jar. Personally, I wouldn't recommend a colonization temp over 80F. When large spawn bags are used instead of jars, stick to 75F. Good luck.
