The Beginning- substrain selection

I wanted to post this into the grow logs but I don't have permission to post in that section of the forums, so here I begin.  I've had a few experiences over the years where I tried various little experiments w/ mushrooms, 2 of 3 being successful on a smaller scale.  I have just recently started up on a new grow and I am glad to say that I have taken it to another level in my own opinion.

This is currently a  joint project going on w/ a fellow enthusiast and general associate.  At present we have 3 pint jars fully colonized- South American, Brazilian, and Thai. Lipa Yai. Along w/ this is a spawn bag that's probably a little over a 1/2 gallon of WBS inoculated with United States Virgin Island, partially colonized quarts of Thai Mystery and Australian, and partially colonized pints of Argentinian as well as Ban Hua Thanon.

While these grains are spawning I also decided to try my hand at agar for the first time and do a little substrain selection.  I used the grocery store agar tek in the tiny half pint jars. On the agar I started Cambodian, Thai Mystery, South American, Brazilian, Hawaiin, Texas, and Puerto Rican.  So far the big winner has been South American which has been surprising.  I'm not sure but I think that it may be due to some slightly adverse conditions that took place in the form of below ideal temperatures during incubation for a short time period.  Regardless the South American has been the hands down winner so far from multiple innoculations.  I'm going to isolate some substrains on the agar and clone some of the nicer fruited mushrooms as well.  Although the South American has been the winner so far I have high hopes for most of these strains given ideal conditions which has been recently fixed(there was previously very little to no incubation) but now we have a full chest freezer at our disposal.  The thing is a good 6' long and 3' deep; it's a welcomed addition to the family:grin:.

Tonight we're inoculating 12 pint jars using mycelium water made from the colonized pint jars (south american, brazilian, thai. lipa yai).  I think I'm also gonna whip up another batch of agar but I have to check out another recipe to make so the cultures stay decent, so I guess it's off to the store tonight.  I might pick up some more lids and stuff while I'm there(got some plastic ones last time and don't like them that much). Almost forgot we just got some spawn bags so we just made up a bunch of them, 12 actually.  It's kind of a test run.  We're seeing which will be better bulk BRF or bulk Grain (WBS/Rye).  We're hoping that the bulk BRF bags do there thing as fast as the grain bags because the BRF bags are alot easier to prep.  I personally like the Rye.  I figure that if I can eat it, they can too.:stoned:  We made up 10 BRF bags and 2 Rye bags all somewhere around a 1/2 gal maybe a little bit more.

The spawn bags that we made up are getting spawned to trays and getting fruited in a green house.  I'm not sure what kind of trays we're going to use yet, I have a bunch of 12 qt sweater box sterilite containers. I have a Vicks coolmist humidifier (from target) and we picked up one of those ultrasonic fogger discs that you drop into water.  The only problem is that our ultrasonic device didn't come w/ one of those floating things that regulate the height of the disc.  This isn't really an issue for now with the gh but it will be in the future when I make one of agars large easy humidifier setups in a 5 gal bucket using giant airstones, industrial airpump, fogger disc, timer, aquarium heater, and squirrel cage fan w/ a couple pieces of pvc.  One thing at a time though(yeah right, lol).

Anyways, that's where I'm at for right now.  I'll have some picture to upload to you guys in a little bit after I get that whole thing figured out.  Other than that I have  a whole bunch of stuff to do so I'm gonna get going.

By the way I wanted to put this in the grow log thread but I don't have the ability as of yet or atleast I don't think I do.  If possible could one of the administrators move the thread at some point it would be appreciated, but if not I know that you guys will be busy and there will always be more to log:stoned:

Thanks man.  I just noticed a little bit of contamination in my U.S.V.I. spawn bag.  It's nothing too major, but I'm not sure what I want to do with it.  I know that I could probably work around it and probably be successful but part of me doesn't even want to mess around w/ it.  I'll have a bunch more bags ready to go in a little bit so I'm thinking it might be best to not have any contams around.  Do you guys think that it is worth it to mess around w/? it's not trich it's a grey contaminant. I'm still a newb so I don't know all of the contams right off the rip and hopefully I won't become all that good at identifying them:stoned:  I'll definitely post up some pics.  We already took some pictures and we'll take some more tonight.

I've got an update for today, nothing too crazy but progress was made.  First off a couple of strains have been dropped from the line up due to a contamination and some slower performers.  Because of this I will not be running Argentinian, Australian, or U.S. Virgin Island.  On the other hand though I made up a bunch more grain and agar today, so hopefully things will be rockin' pretty soon.  'Cause I'm droppin' az couple strains I felt it only right to replace them :stoned: so I'll be starting up some B+, Yosterizi, and Amazonian. 

The agar is going well.  You can tell just by watching it that Puerto Rican is a beast; I have high hopes for PR and Hawaiian.  They are all performing well but I kind of have my hopes set on those two.  Only time and conditions will tell though.

The "new" incubator is holding temperature perfectly and everything seems to be on track except for my sleeping habit:crazy2: . Anyways, I'm kinda tired and just figured that I'd post a few pictures now that I have a little free time.  I should be able to get some good pictures going tomorrow as it's time for inoculation.  I have high hopes for the morning.

Sorry, nevermind about the pics for now, I just tried to load them and realized that I couldn't load .jpg . I'll post some pics up just not tonight, I'm beat.

Nice, look forward to seeing pics of lipa yia, i have some going at the moment, there on cakes, its not going as good as the last cakes to fruit, but atleast i got some going



Its not much, seems to be a little less than the last cakes. But that 1 shroom is not showing any signs of cap opening, im homing that i can keep it growing a bit longer so its extra large and take a clone.

I injected 10cc,s of water into each cake 2day as they seemed to be getting a bit dehydrated. Heres hoping i have a monster shroom to eat!

My Lipa Yai jar is super colonized at this point. It was not the fastest of the strains but it certainly did the most thorough job of consuming the grain. I have high hopes for Puerto Rican, Hawaiian,and Lipa Yai. All of the strains that I'm working with are supposed to be really good so as long as I get through these transfers I'm bound to have a winner of some sort.

Today I'll be knocking up the birdseed and various other spawns that we cooked up during the week. I'm still pretty new and every time I processed the grain I noticed something else that I could do that would improve my results. Because of this we ended up processing 51 1/2 pounds of spawn this week. 5150, the code for insanity; not that 50 lbs is that crazy but it's a decent amount of stuff. I almost went out and bought some more WBS too, I would have ended up w/ 71 1/2 lbs of stuff, lol. As of right now we have about 30 spawn bags give or take; the last 5 of them have to get pressure cooked this morning and then we are good to go.

We have 3 different types of spawn going as of right now: larg BRF cake, rye, and wbs. The large BRF cake was kind of an experiment to see how well it does in comparison to the grains. We were hoping to be able to use the brf because it is easier to prepare in a bulk quantity (from a standpoint of volume not weight)as I am going to use it for inoculation and not fruiting.

That's the next thing that will have to get worked on is the fruiting substrate. We basically took the poo w/ extra's recipe and just bulked it out using variations here and there. My thought process was that so long as the medium is pH and nutritionally balanced w/ the correct moisture content and properties the more complex the substrate the better for the mycelium.

So yah, today I will be taking my first transfers from the agar cultures. I made up like 16 more agar blanks yesterday and I already had a couple sitting around so I'm pretty well stocked now. Aside from the transfers I'm also going to start up another couple of strains today also as mentioned.

I have a bunch of pics that I cropped up that I want to load up on here but they're in .jpg format like I said so I have to go on the internet and see what I have to do to convert these bad boys. I'm hoping that it's something simple, I'm not a computer wiz kid by anymeans.

Well we just made up a whole bunch of mycelium water using colonized pint jars of wbs.  We made Lipa Yai, South American, Brazilian, and Ban Hua Thanon mycelium water.  We are using this water to knock up spawn bags of various spawn media(rye, wbs, brf).  We just got done knockin' up the first couple of bags and need to go to the store to stock up on some stuff. I have to figure out what I need to do to get my pictures up on here; they're in jpg format and won't work.

Already went to the store once today, picked up a bunch of wbs.  I saw a few new teks that made my whole approach to grain preparation alot easier especially when using the grain for spawning like we are, so I did up a little extra, lol  :bongload:

Just knocked up a "couple" spawn bags.  I'm really digging it now that I have someone else to work with.  I've spent alot of time over the last few years going incognito by myself; it's nice to have someone else around. It makes things go alot faster as well, way more than twice as fast. We still have even more grain to knock up tomorrow that I made up quick today on a whim to test out a new tek(new to me), so there's a little excess if there ever was such a thing . :bongload: Anyways I still haven't figured out how to load up these pictures which is really annoying me.  I'm going to do a search on the subject but I don't understand it.  I try and attach files to my post but it says that I can't attach the jpg files.  I don't know if there's just something else that I have to do or if I'm going about it wrong but either way if anyone has any advice it would be greatly appreciated.

Thanks so much man, you have no idea how long I've been trying to get these pictures up on here. I wasn't spending my every waking moment on this problem but you definitely put a smile on my face when I checked the thread. Anyways, thanks again and here's some random pictures that I've got for you guys. I should have some more shots coming up for you guys tomorrow now that I know how to do this. Tomorrow I do my first agar transfer, process some more grain, and whatever else my buddy and I can get done.























Sorry there's not too much of a description going on right now but it's kind of late and the pictures are pretty self explanatory for the most part. If anyone wishes I can go more in depth into different subjects but for right now I'm just going to post these pics for people to get a look at.

Very nice images, I'm so glad I could help!

The only question that really pops up for me is regarding your glovebox there, that locking lid looks identical to the style I'm accustomed to, but I have always found the lid to be a poor seal. The plastic just has too much play in it and it has been a bit of a bother in the past, I'm curious how you dealt with that, if it is even a problem for you. I have switched to putting the hand holes in the bottom of the tubs in order to minimize flexing on the lid during use, I've also used tape in the past on the lids that were particularly faulty in that regard...

I think that it really depends on what type of stuff you are doing.  Some things can be performed in open air and other things are a little bit more touchy.  Although I could get away w/ doing many of my procedures open air I choose to do them in a glove box.  Before I do all of this I make sure to clean the room pretty well.  So w/ the clean room the glove box is just kind of gravy and I don't really worry about the seals.

If you really wanted to fix that problem though I would suggest looking into weather stripping or things of that nature that weather proof homes.  You can find this at most hardware stores and I'm pretty sure walmart even has the stuff.  I personally don't think it's that big of a deal though.

As far as designs go, this g-box was not ideal and it was kind of a last minute fudged item to make due.  I have a larger glove box but the front of it got a hole broken into it so it's currently inoperable.  My current g-box has the holes through the lid and my old one had them through the side with a large hole cut into the lid and a piece of plexi-glass fitted.  The plexi-glass has since been used for another project and the container sits there.  Maybe I'll fix it someday but for now it's going to sit there.  But what I wanted to say is that I personally liked my older glove box better because it was 45  or 50 gallons so it had a lot of room, this new one is too small.  I couldn't find another clear container of that size in any of the stores near me.

Anyways, I'm gonna get going, just figured I'd say thanks again.  I'll have some more pics up tomorrow, oh no, today (it's now 2:47am).  Check back in, I have high hopes for the agar transfers tomorrow.  We'll also hopefully get a grain draining frame set up tomorrow as well.  I think that having a large heavy duty surface area to drain/strain stuff is invaluable to me at this point, I think that it will make a big difference.  It's all about the moisture content.

Just a minor update, the heater that you see inside of that freezer will no longer be used as it's thermostat isn't that great and it's honestly a little too big for such a small, well insulated place. So, we're gonna throw a fish tank heater(mayber 2) inside of a 5 gal bucket and put that in the freezer. The only thing that kinda sucks is that it's a chest freezer so it lets heat out when you open it. Oh well, we'll see how it goes. For right now the stuff is just put in boxes in a room that has the heat turned up. We'd like to be a little more cost effective but you do what yah gotta do.

Here's the new heating setup for the freezer. It is yet to be tested but we're hoping that it works. Worst case scenario we might need to get a bigger fish tank heater or a bigger container for the water.







This is actually made w/ a 7 gallon bucket, not a 5 gallon bucket. Other than that it's just an aquarium heater and a small pump to circulate the water.

I was wondering people's opinions on grain prep techniques. We're using our grain to spawn to bulk substrate. Because of this I've decided to make the grain as much on the dry side as I can and then I knock it up w/ a bunch of mycelium water, usually around 40-80cc per spawn bag which contains probably a little over a half gallon and an estimated 2 lbs dry weight of grain.

I was also wondering if anyone has had any experience with the inoculation guns that they sell at mycosupply. I remember reading a few limited posts on them but I was wondering if anyone out there had experience w/ them.

We're slowly working the kinks out of this entire process. This means building a large screen/strainer(4'x8'), ordering inoculation gun from vet supply place, last but not least we're in the process of figuring out a good way of bulk pasteurization. It would be cool to use the freezer to pasteurize the poo in one of the many ways agar has described unfortunately we only have one freezer. We still might be able to work around this but it's not ideal. For now we're thinking about using 5 gallon buckets to pasteurize the poo as the stores around here aren't carrying foam coolers right now ( a little out of season. We would insulate the containers some how, I'm not quite sure yet. Any suggestions are welcome.

I'd also appreciate some advice on pasteurization. I've read many of the teks and know the specifics of what needs to happen(temps and times required) but I don't know the general of it. I know that I should hold the poo (core temp) in the 160-165 range for an hour, but I want to know what temp water I can dump in to a container to accomplish this. Again I will search this more, but any advice would be appreciated.

I'm on a little bit of a budget so we didn't order the gun from a sponsor's site. I'm usually all about it but this time I found there to be a significant difference in pricing so I steered in the other direction. I won't give any specifics but do a search on the internet for pistol grip syringes, inoculation guns, socorex, and whatever other related terms you find in your searches. The guns are anywhere from $20-$200. We decided to order one that's $60 and do 10ml inoculations continuously fed from an lc w/ a large needle and silicone tubing. These instruments are generally used for mass vaccination of animals, it's a veterinary tool.

I'm planning on using alcohol soaked makeup sponges to sheath the needle that way it has minimal air exposure and is constantly being disinfected w/ alcohol.  You just have to be careful not to get alcohol in your spawn or anything else of importance.  I should mention that I don't have it as of yet, it will be here within a couple of days.

Aside from that I'd like to update this thread w/ some new pics.  We built a screen/strainer out of some scrap wood and screen slapped together.  I won't throw out any numbers but there's a decent amount of grain spread across this screen.  The screen itself is 4'x7' for reference :bongload:

I'll also mention that there's a dehumidifier going right by this and that there sill soon be a fan located next to it for air circulation.

Checked in on things and figured I'd post up a few pictures.  We did the grain to grain transfers only yesterday and they are already blowing up.  Here's some pics of the g2g about 24hrs after transfer. I wish I knew which strain is which off hand but I'm not sure.

Well, it's day 2 since the first round of inoculation and things are going well. The g2g is progressing along at a satisfying pace and the spawn bags are showing their first signs of colonization which I don't think is too bad considering it's only been about 48 hours since inoculation.

Today was a busy day and we got quite a few things done, the biggest thing being the agar transfers. I did transfers of fully colonized agar onto fresh agar; two transfers per culture. There were 7 strains in total, so that meant 14 transfers today. The seven strains are south american, brazilian, cambodian, thai lipa yai, hawaiian, texas, and thai mystery. Here's some pictures of the transfers that took place today.









Sorry that there aren't more pictures and they aren't a little higher quality but I'm running short on time to sleep let alone take pictures.

I went on a little trip so I haven't up dated in a couple days. The following pictures were taken 11/09/07 I just didn't get a chance to upload them. So, here's some pics of some spawnbags and my agar transfers. We experienced a 25% contamination rate with the spawnbags, but we are fairly sure that this is because there was a shortage of syringes and we were using each individual syringe more times than we probably should have w/o completely re-sterilizing them. We also did not have injection sites and just went straight through the bag and taped in the open air of a far from ideal office. Given these laxed conditions I definitely did not feel bad about the contamination rate. We just got an inoculation gun which will solve our whole syringe contamination vector thing.














The fifth and seventh spawn bags from the top are contamed. On a brighter note my first experiences w/ agar are going fairly well. Out of 9 multispore inoculations and 14 transfers I only experienced two contaminations. Although I did experience these contaminations I'm pretty sure that in some cases the spores were actually no good as I had used a couple really old syringes that had been severely beat to try and hold on to a couple strains but everything knocked up with them contamed, this was the case w/ the ban hua thanon. This is absolutely nothing against the strain it's just the syringe that I used for it had been beat over the last couple years and more than a couple moves.

I'm trying my best to stay on top of things, but it's hard.  If I'm here updating, I'm not taking care of everything that needs to be done; If I take care of everything that needs to get done, I'm not updating the thread.  I'm balancing it out I guess; I'll try and keep up with the once a day and try to get a little more thorough.  This is supposed to be agar selection isn't it.  I guess I should give some details.

The first thing that comes to mind is that I poured the agar a little thick and the transfer pieces are way bigger than they would ever need to be(thickness).  So, in the future I will definitely be pouring the agar on the thin side that way I get more agar cultures out of it and it's a little less work for the myc as they would't have to colonize the rest of the transfer before getting to the fresh agar.  I suppose putting the transfer in upside down could solve this problem.  I'm not sure whether the lack of light or air would have adverse effects on the transfer though.  If anyone could chime in on this matter it would be appreciated, whether it be about actual pouring and transfer technique, or about the the matter of putting the transfer in upside down to hasten the time it take to make contact with the fresh agar.

So far, of the multispore inoculations(WBS spawnbags/jars) the Brazilian is the absolute fastest hands down.  The only other strain that is remotely close in colonization time in this experiment here so far is South American, which still does not perform on the level of the Brazilian. As far as the agar transfers go Texas is the standout candidate so far.  Hawaiian, South American, Puerto Rican, and Cambodian are not too far behind but they aren't as far as Texas.  The Thai Mystery is the only one that seems to be behind everyone else as far as agar goes.  With the g2g pint jars both the Brazilian and the South American are almost fully colonized.  I don't know how many days that is exactly but that is a pretty fast colonization time.  The amazing thing is that the Brazilian spawnbags are going to be fully colonized very soon as well.  I must say that this mycelium water technique that I've been using for inoculation is beautiful.  You don't have to wait for LC to colonize; you can essentially control the levels of mycelium present in the water; it's instantaneous so long as you have at the very least a partially colonized jar.  Coupling this mycelium water technique with that new inoculation gun should prove beneficial in overall results and happiness levels, lol  :bongload: One thing that was noticed by my friend is that with the strength of the spring in the gun and the repetition of use, our hands should become fairly strong, lol.  The gun only shoots 10ml at a time and we're prolly gonna be hitting stuff with anywhere from 30ml-80ml.  We would inoculate with more(maybe even will) but it depends on the amount of inoculant available at the time. It shouldn't be too much of a problem though.

The last round of spawnbags that we did we decided to bulk up in size.  The last ones were probably about a third full, while this batch is probably a little bit less than half full.  I want to see how far the limits can be pushed per bag, I read what RR posted on this matter but I figured I'd give it a try myself.

On a similar note has anyone had experience inoculating spawnbags of coir with mycelium culture of some sort(lc, water)?  I read about someone experimenting w/ these techniques to hasten time but I could not find anyone that had documented results.  If this was a reasonable technize I would imagine that the preparation of spawn would become exponentially easier, or so I hope :grin:

I would also like to hear everyones opinions on bulk spawn preparation.  I'd like to focus on prep. time/difficulty and as well as any other inherent pros/cons involved in the various possible spawn media(practically endless).

Anyways, I'm gonna go get my day started and actually go check on everything.  Maybe I'll snap a few shots for you guys as well, I'm fairly impressed with the g2g as well as many of the spawnbags performance.

What exactly do you have mounted in the holes of your glove box it looks like a plastic of some sort. Is that a fan mounted for positive pressure? How's it's overall performance/contam. rate?

So here's a little morning update.  Everything seems to be going well, the braz g2g is just about done, the South American g2g is almost there as well.  Quite a few of the bags are well developed as well.  The first pic is the braz g2g and the second pic is the SA g2g.

I've also noticed that the drier BRF that's closest to the heatbomb seems to be the fastest overall runner.  I think that it's due to the low moisture content of the BRF because wetter/clumpier BRF didn't work anywhere near as well.  That's more my buddy's project than mine though, I like grains.  If the BRF is fastest, easy, and works, I'm all about it though.  Anyways, here's some pics from this morning.

We just got a new pressure cooker in the mail from a friend via kickdown:stoned: We now have 2 working Presto 23 qt pressure cookers.  We have one other one as well but it got bent in an accident and isn't exactly perfect to say the least.  Maybe with a little force we can get it back in working condition.  Either way though, 2 23qt pc's will be awesome.

The inoculation gun is quite the little tool. We haven't used it yet but we tested it out w/ water and it's awesome. I'll also mention that once there is liquid inside of it the gun is much easier to operate/squeeze. We made one slight modification to the gun which was to make a glove that slides over the back barrel because it's exposed and seemed to be a possible vector of contamination. Maybe it's overkill but figured better safe than sorry.

There's also a fridge in the works as well. We already have a large upright fridge/freezer that we are going to be able to use for another incubator we just need to clean it first as it's pretty funky.

More pics for you guys. I'm trying to be a little bit more thorough in my whole procedure here so I'll try my best to give you guys some detail.  I might have messed some of the code up for the pics so I might have to mess with it a little to make it accurate.

Everything is going at extreme rates.  This is due to an accidental increase in incubator temp. that took place bringing it someone in the near 90 degree area.  We suffered three more contaminations of a bags that were touching the giant heatbomb.  The contamination occured directly where the bag was touching the heatbomb so I know that's what caused it.  Kinda sucks, and we prolly could salvage it, but no need to be messing around w/ funky stuff.  So that's twelve bags in total out of 31 that were contaminated.  I have mixed feelings about this.  In all honesty I feel a little crappy if I focus on it(38.7%contam. rate :shake: )but I do know where we went wrong(identified the vectors of contamination and errors in technique) and we have moved on.  Fortunately we had the foresight to prepare for contamination and did a little extra :bongload: So, although it may hurt the pride a little we are still in good shape. It was basically inevitable that we would mess up but I figured that we would play the percentage game, accomodating for up to 50% contamination within our run and still being in okay shape for our goals.  Not to say that we expected 50% contamination but we knew that there would be alot of little kinks to work out of these little systems especially because we didn't follow anyone else's setup verbatum.  Once we get our whole system dialed in we'll be fine, heck, we're still fine right now; pride's just a little hurt. What can you expect though, we are newbs.  I think that we have all the tools necessary to do just about anything you see on these forums, we just don't have the experience yet.  This is the other reason the I changed the threads title over to the beginning.  Like I said though, we're still doing alright

I'm actually thinking about hooking a pump/hose up to the heatbomb and doing a little radiant heating throughout the freezer.  That way we could turn the heat down a little bit and keep it more controlled/even.

As far as the grain goes Brazilian is hands down the winner so far.  It is basically done colonizing the pint g2g that I did as well as almost being done in some of the spawnbags.  :bongload:

Well, all I can say is BRAZILIAN!! :rockon: It is literally tearing through everything that it touches.  I've actually noticed that the Brazilian almost seems to be isolating itself.  The rhizomorphic growth is so fast that it just dominates.  The rhizomorphic growth is not uniform nor singular but it is definitely visible throughout the various spawns that have been started.







The agar transfers are just about ready for their second round of transfers.  It's hard to tell because I can't see into the top,  but I don't think that I have that much sectoring going on.  The majority of the growth looks like a white mass, not rhizomorphic.  There is rhizomorphic growth though on the edge of the culture that start to shoot up the walls of the jar.  Oh well, maybe it'll take me two more transfers instead of one, who knows. Any advice would be appreciated.

One more thing that I would like to say, the inoculation gun rocks, it's amazing.  For any of you that have been thinking about investing in one of these things go for it.  If you think it may be remotely practical for you get it, it cuts inoculation time down so drastically.  So, if anyone is looking for a review, I give the inoculation gun 4stars.  It could have had 5 stars had we sprung for the more expensive version, but this one works just fine for $100 less.

They definitely are tomentose, they were inoculated using mycelium water from a multispore inoculation. This culture has not been worked at all nor has there been any effort made to isolate that particular culture. What I was trying to say and maybe didn't quite say so clearly was the brazilian spawn bags have areas popping up all over the place of pronounced rhizomorphic growth. This is not that significant at this point but it gives me hopes for the future. The pics that I last posted weren't really trying to focus in on the rhizo areas. I will say though that the rhizo area aren't that big and are sporadic but of all the spawn bags going this strain seems to be displaying some of the most desirable characteristics. Again, I know that all of these strains still have to be worked quite a bit, yet another reason why I retitled the original post The Beginning- substrain selection. Either way though, the basic jist of what I'm trying to say is that I'm pleased w/ what is going on right now. Many of my spawnbags are basically fully colonized at this point, 13 days after inoculation. Everything can and does change though; we'll have to see what the future holds.

You could definitely slice off a little hunk of the rhizo and transfer it to agar. From there I would imagine that depending on how carefully you sliced you could prolly have it cleaned up in two transfers. Now this is just a guess as I have not done it but I figured it may give some others a little more perspective.

That's basically what I have going on right now, substrain selection. I've got my eye on different traits and then from there I'm just cutting 'em down. I have 14 first generation transfers from multispore inoculations right now on agar. Those transfers will be getting transferred soon bringing it to the second gen. transfer. I'm not sure what the actual terminology is for these procedures but I think you get what I'm saying.

Honestly after these bags fruit out I'll prolly clone a couple of the fruits as well. We'll see how things go. I'm just enjoying myself.

So far I love the brazilians performance on the grain. South American has been the tried and true performer through below ideal conditions. Hawaiian has very strong growth and I'm very interested in it's bioassay.

I have a quick question to throw out there for everybody concerning wood as a substrate. We have recently gained access to a near unlimited supply of hardwood shavings. My question is about wood preparation as a substrate. I read another members postings mentioning a fermented wood additive in substrate, and I'm wondering if something like this should be done or if they could be used as is after being pasteurized. Anyways, if anyone has any experience supplementing cube's substrate with wood a few pointers would be appreciated. Right now we have a large shopping bag of maple wood shavings that are about 3-4" in length, a mm in thickness, and a cm wide. There's a slight possibility that there is some beach wood in the mix as well, but not much.

It's cool man, I'm pretty certain that it should be fine but I was just wondering if anyone had any suggestions. We're thinking about pasteurizing the wood separately then mixing and repasteurizing. I'm going to look into the whole wood fermentation a little more but for now I'm a little stretched for time addressing the basics.

For the tropical species you're growing, you want to stick with dung or straw type substrates. Wood isn't their choice of food and performance on wood is quite poor.

Well there were a couple of thoughts going into this whole thing. We are not all that limited in space within reason, so we can do a decent amount of substrate. I've seen people fruit this species on wood before, granted it didn't perform as well as h-poo does but I'm not looking to grow on straight wood, just using the shavings to throw into the substrate mix, I figured that having the more complex structured substrate would benefit the mycelium and flushes. Cause the wood doesn't break down that fast I figured it would promote more flushes, kind of a time release fertilizer of sorts.

Is it recommended to just stick to the poo and straw or can adding things to the substrate to vary up the size, shape, density, nutritional value, and other characteristics of the substrate. I don't have it here in front of me to reference but I remember the mushroom cultivator stressing these points, although I think it might have been leaning more towards compost. I figure that even if that were true the same principles would hold true. Honestly I don't know, it's just speculation, if someone with some experience w/ this could chime in it would be appreciated. I understand that RR already posted an answer but I don't know if that's just in reference to cubes on wood vs. poo, where I was talking about poo supplemented w/ wood, among other things.


Edit- After re-reading RR's post I understand a little bit better what was being said. I understand that poo/straw would be a better substrate, but I still wonder about the structure of the substrate being improved by the wood. Either way I'm gonna be using it cause we have the space , it'll work, and it's more substrate.

Sorry for not updating in a couple of days I've been extremely busy trying to get everything done around here. As usual things have changed since the last time I posted. As of right now the two major winners of the agar cultures are Brazilian and Hawaiian, both of which have extremely rhizo growth. I hadn't checked these in a day or so and they got a little overgrown, but hey, they look clean, and they make for good pics.

Sorry I haven't updated in a little while things have been a little off lately between odd work/sleep patterns, injuries, life in general, etc... We ended up taking a bunch of our brazilian spawn bags and spawning them to four 7 gallon mason trays of h-poo/stuff the other night. 3 of the trays are brazilian, one of the trays is south american, and we didn't spawn the lipa yai to trays.

The trays are now incubating in a chest freezer due to low temperatures in there surrounding environment for the time being. The freezer is staying at about 84 degrees and the heater is not plugged in. I figured that I'd check it a couple more times and when it really starts to go I'll drop the temp in the freezer a little more. The freezer holds temp extremely well, it's been at temp for a little over a day w/ no heater after having 4 large trays put into it that aren't warm by anymeans(should have been but weren't, atleast first half wasn't). Either way things seem to be going well and I have high hopes for things. The trays should colonize pretty fast, we spawned them pretty heavy, so hopefully it isn't too long now.


EDIT- Here's a pic of one of the brazilian trays. The trays are approximately 20" x 30" and hold 7 gallons. We spawned two spawn bags (1.5 gals or so) to something like 3/3.5 gals of h-poo, coir, coffee grounds, kelp meal, gypsum, and some other stuff too. We spawned to the trays two days ago and this was the result

I'm seriously impressed to say the least. I think we may have to drop the temp. just a little bit as I can see the tray is sweating in the picture. At this rate it shouldn't be too much longer now, lol.

Did you BRF bags go sour yet?  Been there done that, didn't work for me with the BRF.  Grains are were it's at!

I think this inspired me to check out brazilian!

A couple of the BRF's went sour, but the majority of them worked out alright. A couple of them ended up a little too moist/clumpy and got contamed in the bottom corner of the bag where the seam/fold is. But I had 3 that worked very well. I will say though the grains are definitely where it's at. I'm thinking about getting into g2g. I've done it before, but never on a large scale before.

I'm actually going to do a bunch of agar to grain transfers as I have over 20 fully colonized agar cultures that have one or two small pieces out of them each. That should be fun.

The Brazilian is definitely where it's at. It's definitely becoming the center of attention these days but I keep the other cultures going half for fun and half in hope of finding some bad ass sub strain. Hopefully things will go well, so far, so good.
