What is the purpose of the polyfil in the syringe?

When you input fresh air via the syringe, do you fill up the vacuum? If not - wont the outside pressure be able to push air through the punctured silicone?

What is the purpose of using two kinds of syrups?

I use both for good reason, a) pans dont like karo alone b) both cause quick colonization c) it's how i do things YO.

I dont let off all the vacuum no, i leave it slightly under pressure, keeps contams out.

the silicone is self healing, and no, it doesnt leak. The polyfil acts as a filtration medium

How does under pressured jars keep contams out? I would figure it would make it easier for stuff to get in. Over pressured on the other hand...

its not "under pressured" its underpressure. that means there is pressure being exerted on the medium.

only to the inside of the PC ..
when I make a LC with rubber plugs as inj sites .. they form a vacuum
a needle with polyfil will vent it to normal air . curious on the "pressure" part myself..

less then .5 cc's of spore solution.





so you added less than half a cc and got results like that. thats badass. i was always told to use between 2-3 cc's but if i can get by with only half a cc thats awesome. like i said before great write up man very easy to follow

and yea, you only need about a half cc of spore solution to inoculate the LC.

LC 2 LC transfers about 2 cc's for explosive growth.

Clone flesh takes about 4 days.

agar wedges of isolate rhizo takes about 3 days.

Pans take about 2 days.

sweet tek, ive tried lc a couple times, i dont have a pc YET so i tried to steam sterilze it both times. didnt work. yo jeetered..have an extra pc laying around anywhere :bigblunt:

OK, guys tell me if this would work. Instead of using a syringe with polyfill for the air exchange(s). Can't I just open a new sterile syringe/needle and suck up some air from the laminar flow coming out of my flow hood and inject that. That way I can carefully recap the syringe with a new un-opened sterile needle tip, and use the same syringe in the same manner at a later date for other air exchanges?

I absolutely cannont find malt syrup anywhere locally...I already have the karo and everything else needed. Do you think 2 tbs of karo, in place of 1 tbs karo and 1 tbs malt, will be adequate for a cubensis strain?

This is awesome timing man!! :smile: i've been wanting to start an lc for quite awhile because for some reason my spore syringes keep failing to get the job done.. i've seen no results on 4 innoc. in a row so i want to turn my spore print into a lc. this tek is the easiest one i've read yet and i have everything i need to do it. thanks for the newb friendly post any all the info
