... After a few years hiatus my neighbors cousins stepsons former roommates bird is giving mycology another try. This odd bird made certain to PC it's jars for a good hour and a bit before inoculation. The only equipment missing was a glove box. One jar in particular was pre-sterilized and left a good week before inoculation. (any contams would likely have shown in this time)
Now despite the precaution of flaming the needle before starting and wiping with 70% alcohol between each inoculation (Also re-flamed and alcohol wiped prior to said pre-sterilized jar) it seems there was a green mold contamination rate of 5 of 7 jars including the one known to be contaminate free after sterilization.
My question, does this likely indicate a contaminated syringe, or is an alcohol wipe and double flaming insufficient to prevent this level of contamination? There has been poor contamination levels in the past but that was likely due to lack of a PC and amateur skills at creating syringes.

If you made your own seringes I would check your methods. And where in your birdhouse do you inocculate?

I would say it was the inoculant for sure. Unless you had terrible filters on your jar lids.
Even when using a glove box it's hard to guarantee a clean syringe. There is just way too much room for errors in the printing process and transferring. That's why I like cloning so much, it's much more reliable.. of course working with spores at some time or another will always need to be done but I do think using multispore inoculation for every grow is not doing beginners any favors.

Glove box makes a huge difference ime. I tried without, got 9/10 contams. I tried with right after and got no contams.

I have made over 50 syringes with only two bad ones.A glove box was always used.

Again tho, a syringe can only be as clean as the print it came from.

Thanks everyone for your replies.
This syringe was purchased from a vendor, no names here, I will be contacting them in any event. Though I do think I will be investing in a glove box if it really has made a big difference in results. I imagine it's greatly advantageous to be able to spray through most of the air and wipe down the inside of a closed environment for theses kinds of things. Those foreign spores are tricky little things... They're EVERYWHERE!
Double thanks for the links. Filtering through posts and search results for the gold nuggets is sometimes tedious.

What kind of tape are you using for the lids? I've noticed the only time I ever got contams in the jar was when using mailing tape. Aluminum tape has always worked for me. Also I don't think wiping the needle with a paper towel is necessary (unless you are using lc maybe). Flaming it should be sufficient. Hope this helps.
