green/blue mould - due to bad inoculation

On my last batch, i made up 3 pf tek jars, that i sterilised in my nice new 15psi pressure cooker for an hour.
Well this is what happens when ur inoculations are not sterile enough. I forgot to take a shower, put on clean clothes, forgot to alcohol swap the top of my jars before and after inoc and did not alcohol swab the tinfoil cover.
I believe this is what caused my contamination as my first batch's that i made that was my procedure, even though it was in open air (i complelty bleach cleaned my kitchen on both batchs).
this developed in 1 out of the 3 jars.




Don't skim on your sterile procedures!!!
I am now making myself a nice glovebox out of a 30 litre plastic container. which i will carry out any future inoculations, cloning, printing etc.
The first sign of contam was a very prominant blue spot about 5mm in diameter after about 2 days. Within the next few days the green spores developed along with white mould. It has now been about 7 days and something that i did not expect is happening. It appears that mycelium is growing along with the green mould. after 7 days i would have expected the jar to be colinised with green as a previous contamination did.
I cannot be certain that it is infact mycelium growing. I was about to load it into my pressure cooker, but then thought. Im gonna leave it in an isolated cover and see what happens. in another week i will either pressure cook it, or i may take it outside somewhere and plant it in the ground.

It looks like you have both penicillium(blue) and trichoderma(green) molds. Kitchens and bathrooms are the two worst places to do sterile work. Nice move on making a glovebox. RR
