humidity and growth of mycelium
    #7458861 - 09/27/07 05:19 AM (16 years, 4 months ago)

Hi guys, this is my first time growing these little beauties and i was just wondering if i could have a few points clearing up.
I have inocculated 8 jars, 2 of which have two holes in the lids, two with four, and four with three holes (these were small holes for injecting spores and to allow some gas exchange). i then have a layer of vermiculite as a barrier between the top of the jar and the substrate (organic brown rice flower, water and vermiculite). now, i've put the inocculated jars in a plastic box, in a warm dark cupboard (that was thoroughly cleaned etc). should i leave the lid of the box on (it's pretty airtight) to reduce contaminants, or should i take it off so everything can breathe (it's pretty humid in there)?
also when i injected the spores i may have miscalculated the length of the needle etc, so it's possible that some of the spores were caught up in the lower regions of the vermiculite layer, can i do anything to rectify this?
and lastly when i injected the spores, the needle went down into the substrate, rather than against the glass. is this going to be a problem, or will it just mean that the mycelium will take longer to become visible as it's growing from the inside out? its been five days and there is nothing to see yet.
thanks guys, sorry for the all the questions!

breathe...Or maybe wait a few days after the MYC has gotten a good foothold...then open it up so the MYC can continue actually..

Good luck

Im sure there should be enough air in the box for ur jars to colinise, u will want to check every 4 or so days to check on progress and this will allow air into the box.

Usually with PF tek 4 inoc points are best for quick colinisation, the 2 and 3 may take a little longer to colinise, but should be fine.

As for injecting into the substrate rather than against the glass, this shouldnt cause any problems, it will just take longer to see growth, it will start growing thru the substrate. I have done 1 1/2 pint jar injecting in the middle (although this was a not using spores but a liquid culture) and it turned out fine.

As long as everything was done sterile u will be fine.

As for the one that u think u may have injected into the verm, hopefully it has squirted itself into the substrate, the verm layer has no nuitrients so is unlikely to germinate, (although mycelium has grown in verm sometimes and even perlite!)
The verm layer should only be 1/2 inch deep and ur needles are much longer than that, i alway insert the needle almost all the way.

5 days is a bit soon, specially if the spores were injected into the substrate, wait another 4-5 days and check back on them and something should hopfully start poking thru the substrate. Be patiesnt of forget about them for a few days,

yea impatience is normal..lol...Ive got a nice bed of myc im waiting to colonize im doing the HPOO and coir/straw mix...I had to pasturize but its good after the cars are colonized you can keep adding pasturized substrate to get more mycc...BULK TEKS

My fisrt time at these but ill never go back I got 10 times the amount of jars and I started with 1 jar..lol

oh another thing i forgot to mention, dont worry about humidity yet, humidity is kept within the jar at this point, u only have to worry about that when u birth to ur fruiting chamber.
