Agar Questions

Ok, a few more agar questions.

First off I am really getting a kick out of the agar work and As you can tell, I have it working :wink:





I have about 20 strains going and over 100 plates. I have isolated for rhizomorphic and fast growth and was wondering a few things.

First, when do you start to see sectioning? I have a few plates that are on there 4th transfer and I am still not seeing sectioning.

Example (Numbers indicate lineage 2-1-1-1 would be second streak plate  first transfer, etc.)





The other question is, what is the percentage of sub-strains that will not fruit? I am only making 4 to 6 plates from my initial streak and trying to work them down to sectioning. Will I have enough to find a good strain?

The bottom picture looks like a monoculture. Are these clones? If so, you won't see sectoring. I'd also suggest turning the dishes right side up.

RR, These are from spore not clones. On the second transfer or maybe it was the third, the dish was split in half. 1/2 was fuzzy growth the other was ryzomorphic growth. I only took from the ryzomorphic side as I was looking for ryzomorphic and fast growth.

By the bottom picture I must assume you are talking about 2-1-1-2. 2-1-1-1 is the same texture but is growing faster. What the heck, I may just do both and see how they do :wink:

Yes, you want to fruit them all. Don't just pick one. Save in the refrigerator a part of each dish you inoculate grains with, so when you determine the best performer(s), you'll have a fresh isolate ready to expand.

Kind of pointless to take pictures of the dishes with the agar side up. It is proper to write on the agar side though. It won't move around on you as the cover is rotated, so it's better to mark sections, etc..

Anyways I was curious to know your response to his question about what percentage of substrains are non-fruiters RR.

Very few rhizomorphic strains will be non-fruiting. I always ignore the cottony sectors anyway, so that ups the percentage. I won't even venture a guess as to percentage if you tried to fruit each sub-strain that differentiates.

We're looking for more than fruiting strains though. Average number of fruits per flush, weight of fruits, quality of fruits, resistance to aborting, etc., are all factors to consider when choosing a good strain to keep in master slants.
