ok the thing about agar is that all of the growth happens on the surface. thus you get a perfectly flat plane of growth that allows you to remove selected sections that you want (rhizo sections, uncontaminated sections, sectors in general for isolation.) to transfer to a new dish.
now with brf you don't really have that option. you can only see the surface and you can have contaminates that are below the surface. its close to impossible to do a strain isolation from multispore. since you're cloning and don't have to worry about strain isolation basicly all you will be doing with the brf is colonizing it to see if it's contamed and transfering it another jar. so if you're doing that you would get better results transfering clone tissue dirrectly to grain, lc would be second choice to me the reason being is you won't know if you lc is dirty untill it colonizes and then you inject jars. the grain you'll know is clean if it colonized and has no contams it's just more time efficient and grain lc wil last a lot longer than sugar based lc.
oh and dontever trust the oven tek. it's total bullshit look up agars opinions on it and there's pleanty of science explaining why its actualy less clean than open air inoculation.
