hummm couldn't I just scrap some straight in to a grain substrate like wild birdseed
Ive never took a clone before but I'm sure if I work in a glove box I should be OK
Ill have to refresh my reading on taking scrapings I need the rhizomorph roapy parts so I remember
Ive never actually looked at the inside of a mushroom
Ill have to dissect and cook few to see what I'm scraping for

i believe what blood meant was that the mushrooms you will be using will have been exposed to all kinds of contaminates, the mushroom tissue itself will likely be the source of the contaminate, exspecially since it takes so long for the tissue to germinate and start producing mycelium

Agar would definitely be the way to go, but a small bit of tissue cloned to LC in a GB could possibly work.
However, the live tissue to LC is pretty likely to contaminate.

i haven't used shitaki but i can tell you it works with white button mushrooms from the store. make up an L and use a piece out of te center of te stalk or cap as that is the most sterile part of the mushroom. good luck and post results.

thank you everybody,
all information has been noted
hopefully the inside of the mushroom should be clean but I don't know what the growing conditions was
I think i must trial and error

You'll be miles ahead purchasing a proved culture over trying to isolate a fruiting strain of shiitake from spores. It could take years of trying before finding a strain that will outperform what's available. I've made dozens of isolates from spores of my best fruits, and hardly any have fruited at all, and none have fruited as well as my commercial strains. With enough time, money and patience, it's very possible, but understand the numbers are against you with shiitake, where with cubes, it's rare to get a non-fruiting strain from multispore inoculation. Wow thanks never ending learning for me
I thought all strains from all spores and clones will fruit
I will probably try isolate my own strains tho from a superstore aswel just to give it a go and see and learn and become more experienced,
I will how ever look in to some spores too
I have some shiitake grow pegs I got from ebay ( Plugged In to a dead Stump) but thay wont be ready till next summer as ive just plugged them in few months back " I should of used a peg in rye grain substrate "
Ive successfully got spores from closed cap mushrooms from a supermarket but there white cap mushrooms " Tasty but crap compared to Asian super mushrooms"
Ummmmm Shiitake Fried in butter and fleshy tasty slippery PO-ku mushrooms in sir-fry yummmmmy
