Making an LC out of a syringe of sterile water and a fully colonized jar

Ok, I heard about this recently and I'm going to attempt it later, I have a question though:
What is the best way to prepare a syringe to do this? I would imagine I could boil some water and shoot it in and out, fill it and let it cool, then shoot the boiled water out, flame syringe, and aspirate some sterile or purified water into the syringe. I would then flame the syringe again before shooting the sterile water into the fully colonized jar... how does that sound?
thanks everyone
peace

u can shoot boiling water in and out of the syringe a few times and then leave it in there the last time to cool that will be fine, the water will be sterile because its boiling

sterilize 500 mL of water in a jar using your pc. let it cool...
draw out 60 cc's of water from the cooled sterile water jar... inject 60 cc's of the water with your sterile syringe into the grain jars thats colonized... shake.... inject 120 more cc's water into the grain jar.... shake... draw as much as possible back out and put into the remaining sterile water... shake... BAM! 500 mL sterile water LC that can sit in the fridge for months.
at least thats my interpretation of it.

filling the syringe with boiling water works perfect, however, no need to flame a syringe, ever. simply swab both needle and injection site with alcohol...

What I really need to know is what happens with the syringe in between these two phases. I would think that the inside of the syringe has to be sterilized somehow, right? I mean, I can't just take a syringe that was just used for a multispore a few months ago and suck sterile water up into it and expect it to work, can I?
the reason I ask is because I've never had much luck re-using syringes for LCs and such, and I'd like to learn how to do it properly.

sterilize the syringe then too.. fill it with water and wrap in foil and load in the pc (make sure you have reusable syringes, single use only dont do so well in PCs)...
honestly i haven't sterilized a syringe in a long time i buy mine sterilzed and just open them when needed. i suppose boiling water sucking up and emptying a few times as you described would theoretically clean the syringe, but i'd ask the pros on that one, i've always PC'd or bought new.

I sterilize the water that will be used for the LC syringe. In a separate pan I boil the syringe for at least half an hour. Put a lid on the pot and leave the syringe in the boiling water right up until you're ready to go to work.

Incorrect. Swabbing the needle with alcohol does nothing for the contaminants that have become lodged in the interior of the needle. In addition, look at a needle under a microscope. There's many little holes and imperfections on the surface that alcohol is very likely to miss and thus the contaminant molds or bacteria survive. Always flame before first use, and flame again between each jar to prevent cross-contamination. RR
