WBS not colonizing... LC to blame?
    #7437241 - 09/21/07 03:56 PM (16 years, 4 months ago)

Hello all,

9 days ago I innoc'd 7 quart jars with some WBS. There has been absolutely no growth whatsoever.

I'm unsure what could cause this. The jars were incubated at a consistant 79-81 degrees, they still have that slightly sour soaked WBS smell to them (doesnt smell like bacteria), and the WBS was mixed with a small amount of vermiculite which leads me to believe that it wasn't the water content of the jar either.

So then I began to wonder if I even innoc'ed it with anything. I made a karo LC which developed on the weaker side, and I swirled the jar around to break up the myc before placing it in the glovebox and lysol bombing it and letting it sit for 10 minutes. Is it possible that when I sucked the liquid out of the jar that I didn't get any of the myc, and therefore my jars haven't been introduced to any LC yet?

and if so, are the jars still viable if I made them 9 days ago?

Thank you all for your help

"slightly sour" smell IS bacterial bloom. :smirk:

Sounds like your LC might have been contaminated with bacteria.

You should have been able to see mycelium in the syringe before inoculating the jars.

As i understand it, the slight sour smell comes when soaking the wbs to germinate the endospores... thats how you know the soak is effective.
