Beer soaked cardboard
    #742923 - 07/12/02 09:25 PM (21 years, 8 months ago)

Here is an experiment I made with a variation of cardboard substitue for agar:
- boil beer for a minute to let some alcohol out.
- soak corrugated cardboard in it and put it in glass petri dish
- pressure sterilize
- I sprayed some H2O2 on it before inoculation

And here are the results after 1 week:

Some dishes dried too much during sterilization so next time I will wrap them in aluminium foil.

Psilocybe cubensis Golden Teacher

The interesting thing is that mycellium grows also on the underside:

Psilocybe cubensis Koh Samui
Note that this one was too dry and mycellium grew inside cardboard and popped out on the upper left corner.

Pleurotus djamor
Dish is fully overgrown it only looks a bit thin compared to standard agar cultures. However I prefer this one because this strain on agar prefers to form fruitbodies than spreading around.

Flammulina velutipes
This is a bit too aerial for my taste (probably because cardboard is too dry).

Next time I will try to germinate spores on it. What do you think?

interesting.
It should work best for wood lovers, as the cardboard itself is otherwise wasted/just a medium.

Yes, for cubes it's just a sponge containing nutrients.

EXCELLENT. What else can be said. Beer is an incredibly complex medium, and obviously seems to work.
Congrats, the beer-cardboard agar-substitution will make pure culture accessible to just about anyone [unless you're under 21 ]

However...the cubensis looks like it kind of likes it...maybe not as anything but a vessel for growth, but I could see it being good maybe to inoculate an outdoor cow patty..maybe someone should try it.

if you leave off the h2o2, it will work fine, i have had great success germinating spores on cardboard simply wetted and microwaved, then placed spore side up on peroxide agar plates. the cardboard acts as a barrior from the peroxide in the agar, and once the myc reaches it, it takes off like you wouldnt believe. i had a fellow pm me, trying this out, but he used a dextrose solution to soak the cardboard in, and got contamination. i have had 0 contams using straight cardboard/water

Sound like a good idea. It's better to germinate spores on less nutrient medium to avoid contamination.

I found out that cardboard is drying out very quickly compared to agar. I use Micropore medical tape to seal petris and it seems that it lets too much moisture out.
Agar is hard to beat.

No I think there should be no problem, the only thing is that maybe you will need a filter patch on the lid.

I used this BSC a couple of times and I found that the biggest problem is that it is very difficult to cut wedges out of cardboard. Next time I will cut it to small pieces before sterilization and inoculation.

im curious, what would the next step be. inoculate jars or colonize substrate?

Most neetest thing hey Moe? Lol
Nice experiment.
That would most likely be used to innoculate a jar of grain.
That wouldn`t be enough to spawn unless ofcourse the spawn was a tiny amount and you once again used it to spawn and then again and again.
Too much effort.

IME you can transfer wedges to fresh petris to keep it going or inoculate substrate jars.

How much 3% H2O2 do I have to add to a liter of water to get a good strength for spraying over those petri's?
