advanced glovebox question

not really advanced but the avg question of how to build one

I want to know how do u get the jars from the pressure cooker into the glovebox and from the glovebox to the incubator

if its so sensitve as u have to INNOC INSIDE the GB

isnt it just as APT to get mold spores etc after u open the GB and put them in the incubator?

I read and read and saw nothing of this step
so about all I could think was mist the air with a 80% water 20% bleach before making the transfer

anyone chime in on this?

I wipe the jars with alcohol before I put them in the glove box which also has been wiped with alcohol and once they are inside i spray lysol into the GB and seal up the arm holes (really a still air box) wait a few minutes then get to work wearing gloves wiping alcohol on your gloves and cover your arms with tyvek sleeves.

A glovebox doesn't have to be sterile. It should be as clean as possible, of course, but as long as the air inside is STILL the contams aren't blowing onto your work. I clean my workspace and the inside and outside of my glovebox with alcohol, wipe down the the sterilized jars with alcohol, open the lid of the GB, and put them inside. Also, I only use my GB for agar, culture transfers, and syringe making. Inoculation with syringes is just done on a clean counter.
