Agar question

When I streak my agar, I tend to use the traditional method of a zigzag pattern, rotate 1/3 turn and repeat, etc. They way you are taught to do it in biology. I have seen some people just put the spores in the center of the plate.
Does it make a difference?
This is just in respect to isolating mycelia from a contaminated spore print. After that it is transfers.

Nah, the zigzag method is actually the best way to isolate different strains from contamination. I was taught to do it by rotating the plate 1/4 of the way, to get an extra zigzag out of it, but whatever.

Agreed. If there is a heavy contaminant sporeload on your print, the zig zag method allows you to see individual growths sooner, and then transfer each and every growth as soon as it's found to another dish to grow out. This way, the contaminants will end up on an isolated dish, and the actual mushroom mycelium will also end up on separate dishes. RR

Thanks for the info. That is what I was doing and I have had great success with it. It was just strange that so many were suggesting just putting spores in the center of the dish.
And Just for fun, Here the Blue Meanie isolate I have been working on.

It is food coloring. I use it to ID the type of agar. MDA, MEDA, PDA, etc. Each has it's own color.

Here is another pic, you can see the red, green, and the plane agar color. The plane colored stuff was a test agar and it did not work as well as I had hoped.

The yeast contamination was not a surprise to me. I maintain 20 different strains of yeast and it shows up on a plate from time to time. I just take it as a reminder that I need to be a little more careful with my sterile technique. Base on it's position, I would guess that I let the inoculation loop get outside of the air flow and it picked it up before I streaked the plate.

I have some home brewing yeast. Most is wild yeast that I have caught. Some makes KILLER sour dough bread.

Not anymore. I did years ago. I would like to start up again but who has the time? Right now this one hobby is enough. When I first started I didn't realize how much time you can spend playing with Mycology. It's also weird how deep into you it gets. Whenever I go shopping anywhere I always seem to find myself looking at things and thinking about their applied usage for Mush. Growing.

I brew .. I hope the skills I am learning with edibles and agar (have some oyster spores coming from a member here) I can use to store ale yeasts and take some out make a starter whenever I want rather than ordering new yeasts when I want to brew a batch of beer
