New glove box

Hey folks. I had some free time last week and figured I'd build myself a new glove-box for the myco lab. I have a small space that I use for fungus cultivation and I was in need of a smaller sterile box. The last one was overkill and took up to much space.

Here is a shot of one corner of the room with the new box. The room is exhaust vented and the ceiling insulation is sealed over with 3 mil plastic (meticulously!). Overall it's pretty clean - but not clean enough for microbes.


Here is a close-up of the new box.


Filled with your standard goodies for mycelium culture on agar:


The cover is hinged acrylic with a small plastic knob and a weather seal all around the wood:


The lid is secured at the base with a highly sophisticated latch mechanism:


And the arm holes are just 6" cut-outs for my very clean, gloved hands, covered full-time with a dust shield:

nice
something i've done is get a couple cheap plastic flower pots
cut out plastic bottom with hole saw
and insert in those hand holes,
if gloves are desired
a couple of those hose clamps that screw tight
can hold them on.

i use floor tile adhesive to hold and seal the pot itself
or just wedge it in.

really nice .. I was going to make a flow hood and I got a hepa filter coming but I think its too big and finding a fan big enough for the filter may be problematic .. but thats a nice setup ..I may have to make one just like that , can I ask where you got you parafilm from?

That's a great idea. Cheap, easy to find all the parts and I bet it works well! Right on. If I do go to long gloves I will keep that in mind. For now, clean arms and latex gloves will have to do; with the occasional 95% EtOH spray just to keep things honest. :grin:

Not to shit on your glove box, because honestly i think its pretty kick ass. But isn't wood a contamination magnet. Maybe if you lined the inside of the glove box with some kind of plastic. or maybe sheets of ABS. I mean the entire point of a glove box it to keep contams out so the last thing you want to do is have them growing inside. lol, just a though man but you def did a kick ass job on it.

my thoughts exactly!~

molds will grow off the wood.

I'd cover the wood with something...
either some kind of plastic or paint it with some kind of marine paint...etc.

Yeah man, no doubt a consideration - and shitting on the box is cool with me, as long as its constructive :thumbup:.

My last box (similar design) was lined with plastic and I found that when I sprayed it down with something bactericidal, there would be pooling or droplets that took a while to evaporate because there is no flow in there. So for this one, I decided to go without any covering. My understanding was that wood tends to be fairly benign in terms of contams. Plus, when you spray it down before each use, the EtOH or Lysol or bleach or whatever tends to absorb into the material, leaving a dry surface.

I don't know. What do you think?

That makes sense. I guess our cutting boards do get a tad funky after some time. Maybe a coat of enamel this weekend. Thanks for the input ya'll.

Well like i said. they have this stuff called ABS plastic. You buy it in sheets that are maybe 2ft X 2ft and automotive stores some times, wal mart, or like Lowe's. That way you could just screw that to the wood inside and wouldn't have to worry about pools to dry. Or if you cant find that you could always just use more plexi glass. that stuffs pretty cheap and could easily be cut to size. Just an idea.

Great work... But just be very careful using an alcohol lamp in there.
I have read many entertaining threads of glove box exploding.

But other than that I am jealous of your work area.... plenty of room and very well organized.

Nice design, but I do have to agree with the fellas, wood and towels have no business anywhere near a glove box.

There is a place for flow hoods, and there is a place for glove boxes. and let me tell you, everyone should have a glove box way before they have a flow hood.

IMO, real men love glove boxes. :cool:

Thanks for posting your pics, looks really good. Going through the planning stages for my own glove box and this helps a lot.

A question, whats the general methods for getting stuff from the PC to the glove box? Do you just PC with everything wrapped into something so you can move it safely between the two without risk of contams? I keep trying to come up with a way to get my PC in my glove box and it just doesn't seem feasible.

In regards to the comment above about fire in the glove box, are there things to avoid doing that will decrease your risk of 'accidents'? Obviously using an aresol can with open flame isnt a good idea, but what about after you spray, can you lite up then? I normally just use a 10% bleach solution in a spray bottle, any issues there?

leave everything wrapped in foil as you move it from PC to glovebox, un-wrap once inside & box has been sterilized

It's a small incubator. I usually use it when the indoor temp falls in the winter, mostly for plates. I used to use it all of the time but the temp has been good in the grow space, so I tend to leave them in a glove box. The humidity control sucks on it too.

You guys are absolutely right, I just never liked having plastic in my boxes. Ended up relining it this weekend after all. Haven't had any issues with using a clean towel to cover the slips, but I'm always looking to improve. Any suggestions for that? I'm not interested in using glove inserts.
