In an effort to grow mushrooms of the mexicana A strain, sclerotia that were grown on grass seed for 3 months, were aseptically removed and placed on tap water agar. After one month at 25oC the formation of a single mushroom was observed. There are still many hyphal knots on the agar so perhaps some more will form.
This mushroom grows directly from the sclerotium.

yes, just tap water and agar (1%). This is to provide a source of water without nutrients.

I?m not exactrly sure about the chemistry of agar, but I think it is a polysaccharide complex.
Couldn?t it be that pure agar is nutritive too?

Good point, but I think that agar is essentially useless as a nutrient source. In bacteriological work if you do not add 'food' to the agar you get nothing. Then again, this is a completely different form of life, the mycelium could break agar down, althogh it does not appear to have colonized the agar significantly, epecially given the time it has been exposed to it.

what other info do you have regarding the temp, CO2/O2 levels etc etc?
Have you tried tampanensis? I'd like to see some work like this with them.

I did some searching and found out that most imformation is about bacteria. Most bacteria are not able to digest agar (except for a few exotic marine bacteria). Of ocurse this does not say anything about fungi, but since the mycelium does not grow onto the agar, except for 1 cm around the sclerotia my guess it's that it doesn't contain nutrients (except for some trace minerals from the tap water).

You could, probably should, email a few mycologists and ask them if they know of any fungi which could digest agar alone..

Hmmm, and I thought how the agar liquifies when there is a bacterial contamination is a sign of agar degradation.

Agar is an extract of certain phaeophyta (red algae) species. Afaik, it has no nutritive properties that basidiomycetes in particular are able to digest. I wouldn't be surprised if other fungi (oomycetes, myxomycetes?) would be able to break it down. Heck, there's a couple fungi that are able to live on rubbing alcohol! And you thought you were safe after a thorough alcohol prep :-)

Eden Agar Agar 2.5 gm=2 gm carbohydrates.It is in the form of fiber so it will be low nutritive.What is going on is the shroom must come from the sclerotia,not from the agar.

Psilocybe tampanensis often does the same thing on nutrietive agar: once the mycelium has 'decided' to grow a sclerotium, no mycelial mat will spread over the agar. The sclerotium just collects nutrients, grows bigger and produces mushrooms.

There must be som kind of unknown mechanism which induces the mycelium to form either a mat, a sclerotium or mushrooms but I am totally in the dark about what it is. All I know is that a sclerotium produces more mushrooms than a mat does.

I'm not positive on this but i thing that although polysacarides are technicaly sugars, they are not realdily metabolized by most organisms. I know this to be true at least in the case of artificial sweateners.

aren't starches, eg from rice, wheat, rye etc. considered polysaccharides?
they were the last time I checked.....

all you need to do is add some malt extract to the agar before you pc it . it adds nutrients . you can buy malt extract at a brewing supply store. i used it to isolate the best growing . then add to pc'd rye.
