Scalpels and needles will destroy 3/4 or more of the spores. I've got a microscope picture of a slide I made by scraping the spores onto the slide with a scalpel. Almost all have major damage from the sharp edge.
If you don't have an inoculating loop, you can easily make one with a piece of wire. The idea is to gently transfer the spores with the round part of the wire so you don't cut them to shreds. RR

Don't want to use wire? You can by disposable plastic inoculation loops for about $5.00 per 10. Pretty cheap. Just make sure you get the 10UL size. I've used the smaller 1UL and it's was a pain in the ass to use it without piercing the foil.
I guess it all depends on what your preference is though.

I've found that cosmetic "blackhead removers" make good inoculation loops (no sharp edges) but since they cost about as much a decent inoculation loop it's more about local convenience... especially since my wife bought herself another one after her old one went missing

I use a scalpel but I don't drag it along, I try and lift up bits of the print. Depending on the material it works better or worse though.

take a long paper clip and put tape on the end of it like electrical tape so you dont burn your hand when you heat it up.That is what i use and have had good results.

so you'd jus toss them after every use right? being as that they are plastic, you really wouldnt be able to sterlize them. its still a damn good deal though...
