Found a Maggot in my Sclerotia Jar
    #7320893 - 08/22/07 08:33 AM (16 years, 7 months ago)

this comes from :

http://www.shroomery.org/forums/showflat.php/Cat/0/Number/7196559/an/0/page/0

I found last night a little slug inside my Atlantis Jar while I was inspecting it .

First I couldn´t believe it ...but it was true ... it was moving ...

Like yellowish colour with brown "nose and tail"

The jar was only opened while I was inoculating it in my globebox with two pieces of agar.
It has an air filter with 0,45 micron size ...so ... I don´t know what to say ...

I have seen it again right now but I couldn´t take a picture of it in time .

let´s see some pic of " little brow eggs ... or slug shit" ...I don´t know and some places where it seems that the slug has been eating micelium.

LoL, I take it that suction cup looking thing is teh slug??  No idea what happened there.  You can try and salvage it by transferring to agar if you want, or you can set the slug and mushrooms free outside.

Also, if you have a digi cam, then i assume you can get clearer photos.  Leave the jar on a stable surface when snapping the shot, to decrease shakiness.

nice man. I thought I had a worm in one of my jars one time. Maybe it was. Is it a slug or a snail? Snails seem to have more rouded bottoms as apposed to a long body like a slug

it is not a snail, if it were ...it would be unbeliavable ...how to get in there . It can be a maggot larvae as Workman explained to me . But I still don´t understand how the fuck managed to get in there.

But really I found it weird ... The jar is fully colonized and without signs of contamination, forming Sclerotia ... It has been one month aprox since inoculation.

I will try to be Paparazzi tonight hunting the slug to take a photo...

I have discovered more ... They are where I keep my used coffee grounds.

I have a large plastic box outdoor and there is where I punt my spent cakes or contaminated cakes , I mix them with earth and peat and I use them in pots to plant my weed.

Yesterday I cut my first plant and today I have clean the earth of roots and put it back into the box to re-use it. well I found two big ones ...much more than the others ...maybe they are the same ones ...don´t know but this ones are monsters ... I got rid of this two with a hammer !!! One of them was like waiting to get birthed and was inside this kind of earth ball which you can see in two of the pics ...

Weird ::::::::[image][/image][image][/image][image][/image]

Ha, that mud shell is pretty cool. Those are some sort of grub. Different from the ones in your jar. My turtles would love to munch those fatties!!

another Question...

Yesterday I transferred the remaining Sclerotia into new dishes .

I´ve made around 20 of them. I washed the Sclerotia in water and a bit of bleach.

some of the Sclerotia transferred was wet at the time of the transfer because i washed them and put right inmediately into the dishes.

after one day I can see clearly how the Sclerotia is turning white ..so there is myc starting to grow from them ...but ...i have realised also that some of the stones have moved from their original position when i was labeling and wrapping them.

I can see some sort of ...I can´t find the right word ...sorry for my english ... It should be due to the remaining water with bleach ...

my question is : Should I consider this as a contamination ??
Should I prepare new plates and transfer them again ??

I hop it is clear for you with these pics :

I can't understand what happened from your description, however, I have seen sclerotia knocked around inside a petri dish, after transferring, and you should expect to see mycelium growth wherever the sclerotia rolled on the agar.

I have put the slerotia in the dishes , before transferring I washed them with a mix of water + bleach . they were wet at the time of the transfer and while I was wrapping them they were not stucked into the agar so they were moving " around " . As they have moved around some of the water was " spread " in some parts of the agar , there is where you can see something that looks like a contam.

However , I am not sure if it is a REAL contam or even a problem or simply something that doesn´t matter. If it is a problem , I want to be on time to prepare new dishes and transfer them again .

Thanks for all the help. You will have a place in the Shroom paradise !!!

Where that water is, you may get mycelium growth. So don't toss it out if something begins growing unless it looks like bacteria... like this:

Particularly the one on the left.

these are the worst looking dishes.

the main thing is that most of them belong to the biggest sclerotia stone.

I hope you can help

I can't say it looks good, however, is the watery stuff growing, and has it changed appearance since the first or second day of transfer?

Well WTF ... I will prepare new dishes and transfer again , don´t want to risk my precious stones. I will keep this dishes anyway to see what happens. Its been two days since the slerotia is there , so I suppose that when I transfer it again it will be keep on growing in the old dish as well , toghether with the watery thing ...

The funny thing is that I have started from spores ... and now I have around 25 dishes of this.I would never have thought that i would use such a large amount ...

Well thanks for all the help and have a good weekend

after two days .... very poor success ... most of them present the watery / milky stain again ... so it has been a waste of time and dishes.

I might collect them from the plates , let them dry for one or two weeks and clean again and transfer again to see what happens...

Do you think it is a good idea ??

If you are really eager to save this strain, I would take the largest sclerotia (hopefully large enough for the following procedure) and pat it dry with the paper towel. I would then drop it into a solution of 1/4 isopropyl alcohol (70%) and 3/4 water, and let it sit for 2-3 minutes, then remove the sclerotia, and in a sterile environment, pat it dry with another clean paper towel, keep it wrapped until you're ready to use it, then cross section the sclerotia, and section out a bit of tissue from the center with a scalpel or similar tool, and swiftly transfer to a new agar plate.

Thanks for the answer , I will do that with the remaining pieces.

Some dishes are ok but most of them were contaminated.

I will post some pics in a couple of days when it will show a bit more of growing.
