Grapefruit juice (as well as pummelos, tangelos, and Seville oranges) can inhibit enteric cytochrome P3A4, a cytochrome P450 enzyme in the intestine that helps to break down toxic substances, including drugs.

The reaction between grapefruit juice and the enzymes does not wear off. To get the protection back, you have to wait for the enzymes to be resynthesized by the body, which can take 72 hours.

In addition to grapefruit juice decreasing the breakdown of some drugs, it can also change the absorption of others through interference with two other systems. These are P-glycoprotein and organic anion transporting polypeptide (OATP), which are involved with how drugs get out of the intestine and into the body.

I hate how fucking cheap and corny the special effects for the tripping scenes are. Looks like some dudes got together and dressed up in costumes and shot themselves with a camcorder and applied some video effects :smirk: There were THREE instances of good effects in that fucking movie: 1.) the valet's face stretching when they check into the first hotel 2.) the carpet moving when Duke's online 3.) The hotel clerk's head turning into a moray eel

I believe just as Depp and Del Toro(?) are getting their room right before the bar scene and reptiles is best - the carpet and choppy vision is the best rendition of an acid come up I have seen in a movie thus far, but still...

Well I'm not aware that cyp3a4 does much for the drugs that Duke was doing ( I think benzo's though are metabolized via this route)- it's possible. CYP2d6 is much more important in alot of the drug pathways though, and common valerian and ginko can induce (arguably) with cimetidine, prozac, and others inhibiting.

I think the real reason is that Hunter likes grapefruit. He's got them in alot of his writings that don't mention drugs. I've seen him drink whisky out of grapefruits too, in an interview.

Grapefruits inhibit the activity of cytochrome P450 enzymes of the 3A and 1A groups. Some substances, including drugs, are converted by this enzyme. Specifically DXM. DXM is converted to DXO by this same enzyme, only with different groups. Using grapefruit in conjunction with DXM allows one to metabolize (or absorb if that words is better) dxm before it's converted to DXO, resulting in a more efficient trip.
Users have reported their trips are augmented by quicker come ups and trips in faster duration.
