LC vacuum and tyvek
    #7190916 - 07/18/07 12:32 AM (17 years, 6 months ago)

My friend has an LC jar that he made with a tyvek filter and a rtv port. Will the tyvek allow for enough air exchange for him to such in LC through the port or will he have to find some way to get sterile air into the jar.

It should work fine. I know someone that made Liquid Cultures using tyvek filters. He would use a syringe to suck out the LC with no problems. Make sure you break up the LC a bit though, that way you dont get any clumps stuck in the needle.

You don't need air exchange when making an LC. The mycelium doesn't consume enough oxygen...Gas exchange is necessary in the jars when you knock up substrate and Free air exchange is necessary when fruiting....

I think I understand the question.

Tyvek isnt very permeable.
Store bought silicone is very soft and is leaky once punctured.

With your set-up, I doubt your tyvek will even be a factor when it comes to relieving the vacuum.
New UN-FILTERED air will enter the vessel at the point of aspiration at the same rate that the liquid is being withdrawn so only the slightest vacuum will occur and it will have no effect on your ability to draw up syringes.

Use a sterile environment.

I do all my lc in baby bottles and there is no way for air to get at them and I still usually never ever get a vacuum effect when I go to suck it into a syringe,if your worried about unsterile air getting sucked in just do it in a glovebox:thumbup:
