Getting the most out of an inoculation gun
    #6960068 - 05/24/07 03:38 AM (16 years, 11 months ago)

does anyone have any specialized lids they use on jars for innoc. guns? I need some good techniques for using the gun.... i was thinking about the LC jars in a glove box.. with the tube from the innoc gun going into the box and into the jar.... when one is empty on to the next jar..... can someone help with a good sterile procedure for MASS... let me say it again MASS inoculation!! I've been asked to help design a portabella grow and this one of the only kinks to be worked out..... I guess half gallon jars of LC is the most that can be done as far as mass amounts of LC but info on using the gun to its fullest is needed...... thanks ahead of time

By "gun" I'm guessing you mean a pistol grip syringe. Use it just the same way you would a normal syringe. It's bulkier and clumsier, but if you have a lot to do it might save on the hand strain a bit.

let me share a brainstorm of mine....i was looking at aquarium materials and they had two way all the way to five way gang valves.... i was thinking of having a bottle of alchohol with a tube hooked up to one valve and the others hooked up to LC jars and inbetween jars i pump some alcohol through the system for sterilizing followed by a jar of PC'ed water to flush out the alcohol.... the gangway could handle 3 LC jars an alcohol and a water so its like more ammunition with less reloading.... all of this inside a glovebox on a rolling cart for sterility and portability and maybe something else ending in "ity"

its definitely an advantage..... i just want to use it as efficiently as possible.... i needed tips on lids and procedure.... i've got the theory on this gangway valve but if there is something better i'm all ears... the big deal is quickly cleaning and maintaining sterility inbetween LC jar changes

What exactly are you hoping to accomplish? 5 jars hooked together through aquarium tubing going to what exactly?

Sounds like you should master the basics first before you let your imagination run away from you.

I read in another post to put a makeup sponge on your needle that has been soaked in alcohol. Each time you insert the needle it will be cleaned by the compressing of the sponge. Make-up sponges work best because they have really small pores. Just make sure there isn't too much alcohol on the sponge as to not get any in your jars.

I think his user name was "magdesh" ... just my 2 cents...

5 jars all hooked to the gun though the gang valve..... each valve can be cut on and off ...... i run some alcohol through the gun to sterilize.... a lil sterilized water to flush it out then on to The LC.... the cleaning with alcohol inbetween lc jars would be good in case one was contaminated i'm thinking>

btw .... i have mastered the basics.... i have cult experience and i know how to use this gun already...... so yes i'm needin to use my imagination to improve upon the method.... and no its not running away from me..... i'd say it sounds very feasible..... i'm hoping someone else has an imagination too

Well being a girl has it's advantages I guess...

Go to the dollar store, they come in little wedges or squares and are used for applicating foundation.

My bag says this on it: Cosmetic wedges, Non-latex, Non-toxic, Crumble Resistant, Soft, Absorbent, Durable.

Socorex 10 ml adjustable dose.
Will adjust from 1/2 ml to 10 ml per trigger pull.

I sterilize it in a filter patch bag.

In flowhood.
Add new needle.
Place feed tube in LC.
And, go for it.

Can innoc 30/40 jars or bags quickly.

With bags, swab injection site with alc.
Make a pea sized dot with a sharpie marker.
Where you plan to inject.
Inject & cover dot with Scotch packing tape.
(needle holes are hard to see)

In open air & no flowhood.
I flame needle between injections, with a pencil torch.

Thanks for the tips:thumbup:  I'm gonna use hot glue or rtv on the bags to seal up the inoculation.... when i  used tape they would contaminate after shaking around the innoc spot... after pc'n there would be some wrinkles on the bag so it wouldnt seal up i guess... i'll report back with my future experience

Here's a pictoral done by Una of MushMush in Holland. I'm not sure if this is something like what you're talking about or not, boogertool.

Ok, here's our advanced and automated inoculation method:

An agar culture is transferred to a sterilised blender with sterile water.

The blender is turned on and the agar culture is fragmented into tiny particles.

This mycelium broth is diluted over two bottles with sterilise water.

This is the peristaltic pump that will be used to pump to liquid inoculum into the bags. On top is a device we built, ourselves...

...It is a timer with a foot switch, so that the pump will deliver the same amount every time the pedal is pressed

A silicone tube is fitted with a half syringe that is screwed tight. The tube itself is sterilised in the pressure cooker, The bottle with lliquified mycelium is in the flow cabinet, the tube goes into the bottle, then to the pump, then to the needle.

The bags are first soaked with alcohol, and left to stand for a few minutes. They are then wiped dry with a tissue and the needle is introduced. The footpedal is pressed, and presto, 10-100 ml is squirted in.

The bags are sealed with a hotglue gun.

Growth is fast.

The end result, healthy spawn!

I also use this setup to fill tubes with agar for my tissue culture plants.

I'm actually very interested to hear about your ideas using this 5-way gang valve. I've never heard of it, but it sounds like a neat device, almost turning the inoculation gun into an automatic machine gun innoculator! LOL!

As far as half gallon being the biggest you can go for liquid, I just SEARCHED like a madman for you, and spent a good 45 minutes looking, but to no avail. I KNOW Agar made a post about making LC in 5 gallon jugs. Or at least I think I know he did that!LOL!

I just searched all his main threads, and I couldn't find it, anyone, a little help?

Light and Love,
JD

I don't see why you couldn't. Get one of those 5-gallon water jugs and mix it up right, then knock it up. Takes care of itself.

Of course, there is the eggs/basket theory that would contraindicate this tek.
